Ten isolates of Pithomyces chartarum (Berk. & Curt.) M. B. Ellis were grown on surface-sterilised rye grass at three temperatures, and spore production and toxin content of the spores estimated. There was significantly less toxin produced at 24°c compai ed with 20°c, and for all isolates toxin production was lowest at 28°c. At 28°c there was also a marked drop in sporulation. The application of these results to conditions found in the field is discussed.

Pulsed field gel elec tro pho re sis (PFGE) was used to com pare 59 strains of Mo rax ella ca tar rhalis to evalu ate PFGE for the epi de mi ol ogi cal typ ing of this or gan ism. PFGE-ge ne rated pat terns were com pared with those ob tained by small frag ment re stric tion en zyme analy sis (REA) and species-specific probe hy bridi za tion. The strains used in the study were iso lated from vari ous geo graphic lo ca tions and in cluded proven epi de mi ol ogi cally re lated strains. PFGE yielded more unique pat terns than DNA-DNA hy bridi za tion -30 ver sus 18, re spec tively -but fewer than REA, which gen er ated 45 unique pat terns. Strains that dem on strated the same REA pat tern or DNA-DNA hy bridi za tion pat tern did not al ways dem on strate the same PFGE pat tern. For ex am ple, in 23 epide mi ol ogi cally un re lated strains that shared six REA pat terns, PFGE dif fer en ti ated the iso lates into 12 pat terns. Conversely, strains that dem on strated the same PFGE pat tern did not al ways dem on strate the same REA pat tern or hy bridi za tion pat tern. For ex am ple, in 42 strains that shared 13 PFGE pat terns, REA dif fer en ti ated the iso lates into 31 patterns and DNA-DNA hy bridi za tion dif fer en ti ated them into 16 pat terns. How ever, com pared with REA, PFGE yielded less com plex pat terns that were more eas ily com pa ra ble, and com pared with DNA-DNA hy bridi za tion, PFGE was tech ni cally eas ier.
