Ticks collected in the last two decades from free-living and captive wild animals from 28 municipalities of the Brazilian state of Mato Grosso were identified and tested using molecular methods for the presence of rickettsial agents. A total of 4467 ticks (229 larvae, 1676 nymphs, 1565 males, 997 females) representing 27 ixodid species were collected from 235 species of amphibians, reptiles, birds, and mammals from three different ecoregions (Pantanal, Cerrado, and Amazonia). The species Amblyomma parkeri, Amblyomma romitii, Amblyomma varium and Ixodes luciae are reported for the first time in the state of Mato Grosso. Amongst 538 ticks tested by molecular methods for rickettsial infection, we detected 'Candidatus Rickettsia amblyommii' infecting Amblyomma cajennense sensu stricto and Amblyomma coelebs, Rickettsia sp. strain Atlantic rainforest infecting Amblyomma ovale, Rickettsia sp. strain NOD infecting Amblyomma nodosum, and 'Candidatus Rickettsia andeanae' infecting Amblyomma sculptum. Our results represent an impressive expansion of knowledge on tick fauna and rickettsiae and are essential for understanding the ecology of ticks and tick-borne diseases in the Neotropical region, particularly in midwestern Brazil.
RESUMO.-A ultrassonografia é um método de imagem não invasivo e uma ferramenta importante para o diagnóstico de uma variedade de enfermidades de animais. Neste trabalho foi realizada a técnica de ultrassonografia transabdominal em cinco tamanduás-bandeira (Myrmecophaga tridactyla), dois machos e três fêmeas, pertencentes ao Zoológico da Universidade Federal de Mato Grosso e acompanhada a necrópsia de um animal morto de causas naturais. Os animais do zoológico foram anestesiados com Tiletamina e Zolazepam (Zoletil®) e mantidos em plano anestésico com Isofluorano. Foram realizadas varreduras com transdutor linear (LA332) multifrequencial de 3,0 a 11 MHz do fígado, vesícula biliar, estômago, baço, rins, bexiga e testícu-los. Os resultados obtidos mostram que existem semelhanças entre a arquitetura esplênica, a textura e ecogenicidade hepática, a posição e a aparência ultrassonográfica da vesí-cula biliar quando comparado com a dos caninos. Existem diferenças como localização renal, localização dos testícu-los, espessura da parede do estômago e presença de liquido livre anecóico entre o estômago, baço e rim esquerdo em todos os animais estudados. Ultrasonography is a method of non-invasive imaging and an important tool for diagnosing a variety of animal diseases. In this paper the technique of transabdominal ultrasonography in five anteaters (Myrmecophaga tridactyla), two males and three females, belonging to the Federal University of Mato Grosso Zoo and a necropsy of an animal dead for natural cause were performed. The Zoo animals were anesthetized with tiletamine and zolazepan (Zoletil®) and maintained with isoflurane. Linear multifrequency transducer (LA 332) of the 3.0 to 11 MHz scans of the liver, gallbladder, stomach, spleen, kidneys, bladder and testes. There are similarities between the splenic architecture, the liver texture and echogenicity and the gallbladder ultrassonografic position and appearance when compared to the canines. Differences exist as renal and testes location, stomach wall thickness and presence of free anechoic fluid among the stomach, spleen and left kidney in all animals studied.
Zika virus (ZIKV) was first discovered in 1947 in Uganda but was not considered a public health threat until 2007 when it found to be the source of epidemic activity in Asia. Epidemic activity spread to Brazil in 2014 and continued to spread throughout the tropical and subtropical regions of the Americas. Despite ZIKV being zoonotic in origin, information about transmission, or even exposure of non-human vertebrates and mosquitoes to ZIKV in the Americas, is lacking. Accordingly, from February 2017 to March 2018, we sought evidence of sylvatic ZIKV transmission by sampling whole blood from approximately 2000 domestic and wild vertebrates of over 100 species in West-Central Brazil within the active human ZIKV transmission area. In addition, we collected over 24,300 mosquitoes of at least 17 genera and 62 species. We screened whole blood samples and mosquito pools for ZIKV RNA using pan-flavivirus primers in a real-time reverse-transcription polymerase chain reaction (RT-PCR) in a SYBR Green platform. Positives were confirmed using ZIKV-specific envelope gene real-time RT-PCR and nucleotide sequencing. Of the 2068 vertebrates tested, none were ZIKV positive. Of the 23,315 non-engorged mosquitoes consolidated into 1503 pools tested, 22 (1.5%) with full data available showed some degree of homology to insect-specific flaviviruses. To identify previous exposure to ZIKV, 1498 plasma samples representing 62 species of domestic and sylvatic Viruses 2019, 11, 11643 of 18 vertebrates were tested for ZIKV-neutralizing antibodies by plaque reduction neutralization test (PRNT 90 ). From these, 23 (1.5%) of seven species were seropositive for ZIKV and negative for dengue virus serotype 2, yellow fever virus, and West Nile virus, suggesting potential monotypic reaction for ZIKV. Results presented here suggest no active transmission of ZIKV in non-human vertebrate populations or in alternative vector candidates, but suggest that vertebrates around human populations have indeed been exposed to ZIKV in West-Central Brazil.
RESUMO Estudos indicam, por meio de infecção experimental, que primatas não humanos são susceptíveis à infecção por Neospora caninum. Relata-se um caso de um macaco-da-noite (Aotus azarae infulatus), que apresentou sinais inespecíficos e não respondeu à terapêutica clínica de suporte, evoluindo a óbito, encaminhado em seguida para exame anatomopatológico. Amostras de tecidos foram coletadas e processadas rotineiramente para confecção de lâminas histológicas. Microscopicamente, a principal lesão foi observada no coração e consistia em miocardite necrótica multifocal por protozoário, com a presença de estruturas compatíveis com o estágio de taquizoítos de protozoários dos gêneros Neospora sp. ou Toxoplasma sp. No sistema nervoso central, predominantemente no tronco encefálico, havia estruturas semelhantes às descritas no coração. Os resultados da reação em cadeia pela polimerase (PCR) foram positivos para N. caninum e negativos para Toxoplasma gondii, usando DNA extraído do sangue e dos tecidos. Este relato de caso fornece evidências histológicas e moleculares de que o primata em questão foi susceptível a uma infecção natural, porém estudos devem ser realizados para investigar o real papel dos primatas no ciclo de vida de N. caninum.
The population of wild animals is regulated by many biotic and abiotic factors, and parasites are a biotic factor that affects the dynamic and density of host populations. From 2002 to 2014, 62 wild animals from the biomes Pantanal, Amazon, and "Cerrado" (or Savanna), which died in attendance in the veterinary hospital or have been road-killed, underwent necropsy for parasitological examination. Overall, 36 species of parasites were identified from 24 host species. Among the parasites, the most prevalent order was Oxyurida (29.1%), followed by Strongylida (20.9%), Spirurida (19.4%), Ascaridida (16.2%), Pentastomida (3.2%), Echinostomida (3.2%), Gygantorhynchia (3.2%), Rhabditida (1.6%), Plagiorchiida (1.6%), and Monilimorfida (1.6%), especially nematodes, which have more biotic potential and is more easily adapted to the environment than other classes. The occurrence of endoparasites was observed more frequently in endothermic than ectothermic animals, and herein is reported eleven new host occurrences for endoparasites in wild animals.
