Dissemination of carbapenem-resistant Klebsiella pneumoniae poses a threat to the successful treatment of bacterial diseases and increases the need for new antibacterial agents development. The objective of this study was to determine the antimicrobial activity of carvacrol against multidrug-resistant K. pneumoniae. Carbapenemase production was detected by MALDI-TOF. The PCR and sequencing showed that the blaKPC-2, blaOXA-48, blaNDM-1, blaCTX-M-8 genes were present in carbapenem-resistant K. pneumoniae strains. The polymyxin-resistant K. pneumoniae strain exhibited alterations in mgrB gene. The antimicrobial activity of carvacrol was evaluated in vitro using broth microdilution and time-kill methods. For this, carbapenem-resistant K. pneumoniae and polymyxin-resistant strains, were evaluated. The in vitro results showed that carvacrol had antimicrobial activity against all isolates evaluated. The survival curves showed that carvacrol eradicated all of the bacterial cells within 4 h. The antimicrobial effect of carvacrol in vivo was determined using a mouse model of infection with Klebsiella pneumoniae carbapenemase (KPC). The treatment with carvacrol was associated with increased survival, and significantly reduced bacterial load in peritoneal lavage. In addition, groups treated with carvacrol, had a significant reduction in the total numbers of white cell and significantly increased of platelets when compared to the untreated group. In vivo and in vitro studies showed that carvacrol regimens exhibited significant antimicrobial activity against KPC-producing K. pneumoniae, making it an interesting candidate for development of alternative treatments.
Hematological disorders detected in dogs infected by ABSTRACTA retrospective review of hematological reports of nine dogs detected with Hepatozoon canis infection by microscopic examination of blood smears in a laboratory in the municipality of Dourados, Mato Grosso do Sul, Brazil was conducted. This study aimed to evaluate the hematological profile of these infected dogs, in addition to the occurrence of coinfections with other agents that infect blood cells, since studies concerning canine hepatozoonosis in Brazil are scarce and there are some divergences regarding H. canis infection that still require a resolution. The nine cases of H. canis infection were identified among all dogs examined at the studied laboratory in 2009 and 2010, with an occurrence of 7/1,192 (0.59%; 95% CI 0.15 -1.02%) positive dogs in the first year and 2/1,313 (0.15%; 95% CI 0.02 -0.55%) cases in 2010. The analysis of the hematological reports showed an occurrence of coinfection between H. canis and other agents in two (2/9; 22.22%; 95% CI 2.81 -60.00%) dogs, one with E. canis and another with Babesia spp. (1/9; 11.11%; 95% CI 0.28 -48.24%). Only the blood test of one dog had no alterations, based on reference values. Anemia was the most frequent hematological alteration (6/9; 66.67%; 95% CI 29.93 -92.51%). Although the occurrence of H. canis infection was low, significative hematological alterations were observed in most infected dogs. Coinfection with Babesia spp. and E. canis was detected in two dogs and the hematological alterations cannot be attributed exclusively to H. canis in these animals. Longitudinal studies would be of fundamental importance to determine the causality of these alterations. These results highlight the importance of differential diagnosis in dogs when there is clinical suspicion of infection by hemoparasites, since the hematological changes in dogs infected by H. canis are quite variable.
The rearing environment of pigs can cause a high level of stress due to the lack of stimuli and the impossibility of carrying out natural behaviors. Music therapy is a way to enrich the environment and promote stress relief. Few studies in swine using environmental enrichers focus on functional benefits, such as stress resilience, improved biological functions, or mental status. The effect of environmental enrichment on neurobiological processes is particularly poorly understood in farm animals. Thus, our study sought to elucidate the influence of music in piglets exposed to music therapy in the intrauterine and extrauterine phase on neuroplasticity, evaluating the levels of brain-derived neurotrophic factor (BDNF). Behavioural responses were also evaluated using fear tests related to stress resilience. The productive performance of these piglets was analysed to relate the possible reduction in stress levels to greater productivity gains. Forty-eight sows were used at 90 days of gestation until the weaning of their piglets. In the gestation phase, the sows were divided into two treatments: control (without music therapy) and music (with music therapy). In the farrowing/lactation phase, the sows were separated into four treatments: control-control (no music in any phase); control-music (music only in farrowing/lactation); music-control (music only during pregnancy); and music-music (music in both reproductive phases). Music therapy did not cause a difference in the BDNF levels of piglets at birth. However, piglets born from sows of the music-music treatment did not show a reduction in BDNF between birth and weaning, unlike the other treatments. Exposure to music in the last 1/3 of pregnancy and farrowing/lactation improved the weight of piglets at birth and at weaning. Musical enrichment during pregnancy and lactation was able to cause changes in the piglets’ neuroplasticity and improve their productive performances.
O ovo é um alimento com alto valor nutricional, contendo muitas proteínas, vitaminas e minerais, e com baixo custo o que estimula o consumo. Por ser um produto perecível é necessário armazená-lo corretamente para que este produto possa estar viável para o consumo por longo período. O objetivo desta pesquisa foi avaliar a qualidade interna de ovos brancos e vermelhos armazenados em temperatura ambiente e sob de refrigeração durante 25 dias de armazenamento. Os ovos brancos e vermelhos armazenados em temperatura ambiente apresentam perda de viabilidade aos 15 dias de estocagem, já os ovos armazenados em geladeira apresentam perda de viabilidade com 25 dias de estocagem. A viabilidade dos ovos brancos armazenados em geladeira ajusta-se a uma equação quadrática. Estes dados revelam que esses ovos não apresentam qualidade para ser consumido. Os ovos vermelhos quando estocados em geladeira apresentam-se viáveis durante todo período analisado. Comparando os ovos brancos e vermelhos estocados a temperatura ambiente, observa-se que os ovos brancos adéquam-se a uma equação cúbica, com baixa viabilidade aos 15 dias. Conclui-se que o armazenamento deve ser realizado em geladeira independente da coloração da casca dos ovos.
