A Simple UV-Spectrophotometric Q-Absorbance ratio method has been developed for simultaneous determination of Roflumilast and Salmeterol in synthetic mixture. Q-Absorbance ratio method uses the ratio of absorbances at two selected wavelengths, one which is an isoabsorptive point and another being the λ-max of one of the two drugs. Roflumilast and Salmeterol show an isoabsorptive point at 257 nm in methanol. The second wavelength used is 252 nm, which is the λ-max of Salmeterol in methanol. The two drugs follow Beer-Lambert's law over the concentration range of 2-14 µg/ml. The method was validated according to ICH guideline in terms of linearity, accuracy (recovery study), precision (repeatability, intraday and interday variation), limit of detection and limit of quantification. All the validation parameters were found to be within acceptable limits. The method was found to be rapid, cost effective, accurate and precise therefore be utilized for the routine analysis of both the drugs in synthetic mixture.
