All identified alleles in this study were able to classify the cattle population into three clusters i.e. Bos taurus cluster (Simmental Purebred, Simmental Crossbreed, and Friesian Holstein cattle); Bos indicus cluster (Sumba Ongole, Ongole Grade, Madura, Pasundan, and Pesisir cattle); and Bos javanicus cluster (Banteng and Bali cattle).

Identification of phenotypic and morphometric characteristics in indigenous cattle is important in characterizing breeds of animals. This study was carried out to identify the phenotypic and morphometric characteristics of 813 indigenous Pasundan cattle from West Java Province of Indonesia. The coat color of Pasundan cattle is mainly solid reddish brown (>90%) with black color on hoof (>90%), switch of tail (>80%), eyelid (>90%), muzzle (>90%), horn (>90%) and whitish color on mouth lash (>80%) and reddish brown color on legs (>60%) and rump patch (>60%). The physical characteristic of Pasundan cattle mainly is the hump in males (65.23%) or humpless in female (74.91%), small dewlap size (>60%), presence of back line (>60%) and horned (>80%) with upper curved horn (>90%). The average of withers height (WH), body length (BL), chest girth (CG) and scrotal circumference (SC) of adult male Pasundan cattle were 128.94+16.69 cm; 128.72+15.98 cm; 159.36+15.84 cm and 17.68+3.51cm respectively. Therefore, the average of WH, BL, and CG of adult female Pasundan cattle was 122.56+11.40 cm; 115.63+19.06 cm and 140.13+12.16 cm respectively. Natural Increase (NI) and calf crop values were 18.46% (low category) and 20.40% respectively. Net Replacement Rate (NRR) values were 300% (male) and 507% (female) and indicated that West Java province was still capable to produce seed stock per year.
ABSTRAKPenelitian ini bertujuan untuk mengidentifikasi genotip gen growth hormone (GH) pada sapi Aceh yang diseleksi di Balai Pembibitan Ternak Unggul -Hijauan Pakan Ternak (BPTU-HPT) Sapi Aceh Indrapuri. Empat puluh satu ekor sapi Aceh terdiri dari 21 jantan dan 20 betina digunakan dalam penelitian ini. Ekstraksi DNA genom menggunakan darah sesuai metode Sambrook et al. (1989). Analisis Polymerase Chain Reaction -Restriction Fragment Length Polymorphism (PCR-RFLP) dan sekuensing digunakan untuk melihat situs restriksi MspI pada gen GH. Berdasarkan hasil sekuensing diketahui bahwa semua sampel sapi Aceh bersifat monomorfisme. Frekuensi genotip TT dan alel T masing-masing sebesar 1,00. Mutasi transisi C (cytosine) menjadi T (thymine) pada posisi 1548 bp menyebabkan situs restriksi MspI tidak ditemukan. Fourty one cattle consisting of 21 male and 20 female cattle were used in this study. The genomic DNA was extracted from blood using Sambrook et al. (1989) methods. Polymerase Chain Reaction -Restriction Fragment Length Polymorphism (PCR-RFLP) and mehod of sequencing was used to detect MspI site on GH gene. Based on sequencing results, it can be concluded that all cattle were monomorphism. The frequency of TT genotype were 1.00 and same as T allele frequency. The transition of C (cytosine) into T (thymine) on 1548 bp caused the lost of restriction site.

Short Communication: Novel Single Nucleotide Polymorphisms (SNPs) in the 5'UTR of Bovine HeatShock Protein 70 (bHSP70) Gene and its association with Service per Conception (S/C) of Pasundan cattle. Biodiversitas 19: 1622-1625. Heat stress in the livestock reduces reproductive traits, including of service per conception (S/C). The influence of heat stress inlivestock can be handled through molecular selection. One of the candidate genes that is affecting heat stress tolerance is Bovine Heat ShockProtein 70 (bHSP70) gene. This research was carried out to detect the single nucleotide polymorphism (SNPs) in the 5'UTR of bHSP70 genefrom 44 heads of Pasundan cows at breeding station (BPPIBT-SP Ciamis, West Java). Research showed that 18 SNPs were detected withsequencing method. The insertion mutation was detected in all animal studied and occurred at position between g.1112 and g.1113. Themoderate of polymorphic informative content (PIC) were detected in two SNPs of g.1117G/A and g.1125A/C. Preliminary analysis showedthat haplotype of two homozygote genotype combination from both SNPs had better of S/C value of than the others (P<0.01). Moreover, theaverage of S/C value in heterozygote genotype seems higher than homozygote genotype. However, the further research for clarification ofthis research results through large number of observation is important. It was concluded that two SNPs of g.1117G/A and g.1125A/C arepotential as marker-assisted selection (MAS) for reproductive traits in Pasundan cows because of moderate PIC value.
