Previous studies demonstrated that cGAS pathway is related to the inflammation amplification in a variety of autoimmune diseases. Lysine acetyltransferase family (KATs) can regulate the nuclear transcription or cytoplasmic activation of cGAS through different mechanisms. However, its role and related immunity patterns in systemic lupus erythematosus (SLE) have not been explored. In this study, RNA-seq and scRNA-seq profiling were performed for peripheral blood mononuclear cells (PBMCs) from patients with SLE. R packages were used for bioinformatic analysis. Cell culture, RT-PCR, western blotting, immunofluorescence, immunohistochemistry, and ELISA were used to explore gene expression in vitro or clinical specimens. Plasmid transfection and mass spectrometry were used to detect protein modifications. Eight acetyltransferase and deacetylase family members with significantly differential expression in SLE were found. Among them, KAT2A was abnormally upregulated and positively correlated with disease activity index. Further, KAT2A-cGAS pathway was aberrantly expressed in specific immune cell subsets in SLE. In vitro studies showed KAT2A modulated cGAS through increasing expression and post-translational modification. Our research provides novel insights for accurately positioning specific immune-cell subgroups in which KAT2A-cGAS reaction mainly works and KAT2A regulation patterns.
Short selling mechanism was introduced in Chinese market since March 31, 2010 and has developed for five years. The relationship between margin trading and pricing efficiency is worth studying. In this paper, we focus on the expansion event of underlying stocks happened on September 16, 2013. By using event study method and panel data results, and taking the underlying stocks and other stocks before and after being allowed to the margin trading as the research objects, we reach the following conclusions: First, short-selling constraint makes stocks overvalued and hinders the response speed to messages; second, after introducing the margin trading, especially we are well into short-sale era, not only does the degree of overestimation of underlying stocks decrease, the response speed of stocks to messages improves a lot. Meanwhile we find that there exists a link between pricing efficiency and exchanges, turnover rate, circulation value.
Because it provides favorite ancillary support in terms of technology research and tools test, full-scale simulator for Drilling and Producing is still the focus for overseas researches. The test capability for full-scale simulator represents the level of development for national petroleum exploration and development. That the increasing efforts on unconventional oil & gas and deep water exploration brings new challenges to theory and technology for oil-gas exploration, so the full-scale simulator provides an intermediate bridge for theory and technology innovation. Following the tracks of the development of full-scale simulator, the overseas and domestic classify and development of full-scale simulator was analyzed, and identifying the main technical bottlenecks faced during it establish. This paper describes the development of key technologies for wellhead loading system, wellbore simulation technology and test monitoring system. On this basis, the research prospect for the full-scale simulator was forecasted.
