A sensitive and rapid method using HPLC-MS/MS was developed for the determination of eight glucocorticoids residues in chicken muscle simultaneously by Turbo Flow. The eight glucocorticoids were prednisone, prednisolone, hydrocortisone, methylprednisolone, dexamethasone, betamethasone, beclomethasone and fludrocortisones. Samples were extracted with ethyl acetate and on-line cleaned up through a Turbo Flow solid-phase extraction column without time-consuming pretreatment before HPLC-MS/MS analysis. Sample pretreatment conditions, Turbo Flow conditions and mass spectral parameters were optimized and obtained eight glucocorticoids calibration curves. These curves showed good linearity over the concentration from 0.2 μg/kg to 50 μg/kg with an average recovery from 71.63% to 117.36%. This method could be applied on real samples and provided simple, rapid, sensitive and highly selective analysis, which made it feasible to be adopted in food inspection organizations or carry out quantitative analysis for other banned substance.
One of the main purposes for which dandelions are cultivated is to derive phenolic acids from their processing. Phenolic acids, which are one of the main useful compounds in dandelion, constitute one of the important groups of therapeutically significant bioactive compounds in traditional Chinese medicine. To carry out a relatively full evaluation of dandelion quality, it was found advisable to use multiple indices to avoid conflict with the single evaluation index stipulated by China Pharmacopoeia. Thus, a quality evaluation method was created based on traditional Chinese medicinal theory and relevant statistics on phenolic acids. Firstly, four main kinds of phenolic acids – caftaric acid, chlorogenic acid, caffeic acid and cichoric acid – were chosen as the main indices for quality evaluation through the optimisation of high performance liquid chromatography detection conditions and fingerprint comparison analysis; then, the content of each component was divided into five levels based on the descriptive statistics of 578 dandelion samples and references; finally, the equal weight average method was used to convert content levels of the four components into a comprehensive quality index, which served as the means for dandelion quality to be identified and segregated into grades, as follows: Grade 1 (super high, probability of 0.8%), Grade 2 (high, 18.72%), Grade 3 (medium, 37.28%), Grade 4 (qualified, 32%) and Grade 5 (low, 11.2%). This method is the first to comprehensively evaluate dandelion quality for setting an industry standard in China, and has practical and scientific characteristics.
This paper analyzes the sugar components of different Jerusalem artichoke (Helianthus tuberosus L.) tubers and their leaves. The results showed that the total sugar content of Jerusalem artichoke tuber accounted for more than 70% of dry weight. The content of inulin reached 77.5% on average, and the highest is "Binyu no.1" tuber. The content of inulin in the stem leaf was more than 11.73%, reaching an average of 12.93%. And the effect of inulin was then tested by functional test on constipation model mice. ICR male mice were randomly divided into blank control group, model control group, positive control group, low dose inulin group, medium dose inulin group and high dose inulin group. After 7 or 15 days of continuous inulin infusion, we observe the small intestine movement, initial stool time and amount of black stool. The results showed that inulin had a significant effect on constipation in mice. Finally, we analyse 27 varieties of Jerusalem artichoke by experiments, and select 6 varieties with high efficacy.
