The aim of the work is to develop the industrial technology of tablets with cardioprotective action based on the domestic raw material of cultivated parsnip herb thick extract (CPHTE). Materials and methods. Experimental samples of mixtures of the plant substance CPHTE (Pastinacae sativae herbae extracti spissi) with excipients approved for medical application were used as objects of the study. Results. The results of experimental studies were used during the development of a block diagram of the technological process to produce tablets with cardioprotective action based on the plant raw material CPHTE and an apparatus scheme was tested in the industrial conditions of LLC “Pharmaceutical Company “Zdorovye” (Kharkiv); the control technological parameters for each stage of the technological process of medicine preparation were established. Conclusions. The technology to produce cardioprotective tablets based on the domestic raw material CPHTE was theoretically substantiated and experimentally developed, as well as a block diagram of the technological process of their production and an apparatus scheme tested in industrial conditions by the LLC “Pharmaceutical Company “Zdorovye” (Kharkiv). The results of the experiment were used while development of projects of quality control methods and technological regulations for the developed medicine in the form of tablets under the conventional name “Pastinocard”.
In recent years, herbal remedies gain the increasing popularity in the pharmacotherapy of many diseases. The number of people suffering from chronic pathology of the kidneys has steadily increased today. Phytopreparations would be useful for the pharmacological correction of chronic kidney disease. They can reduce the inflammatory process in the kidneys and, thus, maintain a physiological balance for the implementation and normalization of organism functions. This work aimed to study the anti-exudative activity of effervescent tablets with spissum extract from Betula leaves on a model of carrageenan-induced inflammation and their diuretic activity against in water test in rats. Tablets with spissum extract from Betula leaves were developed at the Department of Technologies of Pharmaceutical Preparations of the National University of Pharmacy. «Canephron® N» (Bionorica, Germany) and «Ortophen-Zdorovje» were chosen as the reference drugs. The study was carried out on 42 sexually mature rats weighing 180–200 g, which were kept on a standard vivarium diet at the Research Institute of Applied Pharmacy of the National University of Pharmacy. Inflammation was caused by the introduction of a 1% carrageenan solution into the hind limb of rats. The study of diuretic activity was carried out using the water test. The rat foot volume significantly decreased during 2, 3, and 4 hours of the experiment under the influence of the studied tablets compared with the control pathology. The reference drug «Canephron ® N» showed a less pronounced anti-inflammatory effect and was inferior in its activity to effervescent tablets with spissum extract from Betula leaves with the same tendency of anti-inflammatory action. As shown by the results of studies, 2 hours after the introduction of effervescent tablets with spissum extract from Betula leaves, diuresis was significantly different from the intact control. Diuresis was significantly higher after 4 hours in the group of animals receiving the studied drug. Similar data were shown by the drug «Canephron® N» by the end of the experiment. For the first time, a study of the antiexudative and diuretic activity of effervescent tablets contained an extract from Betula leaves was carried out. It has been experimentally shown that the antiexudative activity of the developed drug was 28.9%, exceeded the similar activity of the phytopreparation «Kanefron® N» – 22.3%, and was inferior to the reference drug «Ortofen-Zdorovje» – 44.8%. It was found that the investigated drug exhibits the most pronounced anti-exudative properties during the first 2 hours of the experiment. It was determined that after 4 hours the diuretic activity of effervescent tablets with a spissum extract from Betula leaves was 127% and 124% for the drug «Kanefron® N», respectively.
Urolithiasis and inflammatory diseases of the urinary system are of great importance in recent times. Only urolithiasis affects about 15 % of the population. Therefore, the development of new domestic drugs for the treatment of urinary system disorders is an urgent task. In the pharmaceutical market of Ukraine, there are medicinal products of plant origin that exhibit antispasmodic, antiseptic, anti-inflammatory, diuretic effects, as well as dissolve concrements and eliminate them from urine. Aim of the work. To determine the optimal composition of antiadhesive components for the production of effervescent tablets with a thick extract of silver birch leaves. Materials and methods. During the experiment, the effects of macrogol 6000, macrogol 4000, glycine and fumaric acid on the technological characteristics of the model granulates and the resulting tablets were investigated. In the experiments, modern equipment and techniques were used to study tablets ejection force, granules fluidity, strength of tablets, disintegration of tablets. Results. Macrogols of 4000 and 6000 showed the best results in all studies. With satisfactory flow rates, they showed a high anti-adhesive ability and the resulting tablets had the highest rates of crushing resistance. Macrogol 4000 has the most optimal anti-adhesive properties, while the most intense reduction in the pressure of pushing tablets from the matrix is observed at its content under 4 %. Macrogol 6000 has satisfactory lubricating properties, but at a concentration of 6 %. Thus, based on the performed studies, it has been found that for the production of effervescent tablets with a thick extract of silver birch leaves it is advisable to use the macrogol 4000 (or macrogol 6000) as the antiadhesive component. Conclusions. Hydrophilic lubricants (macrogols 4000 and 6000, glycine, fumaric acid) were used to determine the optimal composition of anti-adhesive excipients to produce effervescent tablets with a thick extract of silver birch leaves, among which the best results for all were shown by macrogols 4000 and 6000. According to the results of the research, as the antiadhesive component for the preparation of tablets with thick extract of silver birch leaves, macrogol 4000 was selected which showed high antiadhesive ability and the obtained tablets had the highest indices for resistance to crushing, while the most intense reduction of the pressure of pushing out tablets from the matrix was observed at macrogol 4000 concentration up to 4 % in tablet mass.
