We characterized 208 human Salmonella isolates from 2006 to 2007 and 27 human Salmonella enterica serovar Typhimurium isolates from 1987 to 1993 from Henan Province, China, by serotyping, by antimicrobial susceptibility testing, and, for the most common serovars, by pulsed-field gel electrophoresis (PFGE). The most common serovars among the 2006-2007 isolates were S. enterica serovar Typhimurium (27%), S. enterica serovar Enteritidis (17%), S. enterica serovar Derby (10%), S. enterica serovar Indiana (6%), and S. enterica serovar Litchfield (6%). A high percentage of the isolates were multiple-drug resistant, and 54% were resistant to both nalidixic acid and ciprofloxacin. Of these, 42% were resistant to a high level of ciprofloxacin (MIC > 4 g/ml), whereas for the remaining isolates, the MICs ranged from 0.125 to 2 g/ml. Five isolates (2%) were ceftiofur resistant and harbored bla CTX-M14 or bla CTX-M15 . With the possible exception of the quinolones and cephalosporins, the 1987-1993 S. enterica serovar Typhimurium isolates were almost as resistant as the recent isolates. PFGE typing of S. enterica serovar Typhimurium showed that the most common cluster predominated over time. Two other clusters have emerged, and another cluster has disappeared.
Citrobacter freundii is an infrequent but established cause of diarrhea in humans. However, little is known of its genetic diversity and potential for virulence. We analyzed 26 isolates, including 12 from human diarrheal patients, 2 from human fecal samples of unknown diarrheal status, and 12 from animals, insects, and other sources. Pulsed field gel electrophoresis using Xba I allowed us to divide the 26 isolates into 20 pulse types, while multi-locus sequence typing using 7 housekeeping genes allowed us to divide the 26 isolates into 6 sequence types (STs) with the majority belonging to 4 STs. We analyzed adhesion and cytotoxicity to HEp-2 cells in these 26 strains. All were found to adhere to HEp-2 cells. One strain, CF74, which had been isolated from a goat, showed the strongest aggregative adhesion pattern. Lactate dehydrogenase (LDH) released from HEp-2 cells was evaluated as a measure of cytotoxicity, averaging 7.46%. Strain CF74 induced the highest level of LDH, 24.3%, and caused >50% cell rounding, detachment, and death. We named strain CF74 “cytotoxic and aggregative C. freundii .” Genome sequencing of CF74 revealed that it had acquired 7 genomic islands, including 2 fimbriae islands and a type VI secretion system island, all of which are potential virulence factors. Our results show that aggregative adherence and cytotoxicity play an important role in the pathogenesis of C. freundii .
The human gut microbiota is an important environmental factor for human health with evolutionarily conserved roles in immunity, metabolism, development, and behavior of the host. Probiotic organisms are claimed to offer several functional properties including stimulation of immune system. The purpose of this study is to investigate the effects of a probiotic supplementation on adult volunteers who have contracted the common cold four or more times in the past year. This study is a single center, double-blind, randomized, controlled, prospective trial. Subjects received a probiotic drink containing Lactobacillus paracasei (at least 3 × 107 colony forming units (CFU) ml−1), Lactobacillus casei 431® (at least 3 × 107 CFU ml−1) and Lactobacillus fermentium PCC® (at least 3 × 106 CFU ml−1) or an identical placebo without probiotics for a 12-week study period. The consumption of probiotics significantly reduced the incidence of upper respiratory infection (p < 0.023) and flu-like symptoms with an oral temperature higher than 38 °C (p < 0.034) as compared to the placebo group. Subjects that consumed probiotics demonstrated a significantly higher level of IFN-γ in the serum (p < 0.001) and sIgA in the gut (p < 0.010) as compared to the placebo group and a significant higher level of serum IFN-γ (p < 0.001) and gut sIgA (p < 0.001) as compared to their baseline test results. In contrast, there were no significant differences in the serum IL-4, IL-10, IgA, IgG or IgM between the probiotics and the placebo groups. Results of this study demonstrated that probiotics were safe and effective for fighting the common cold and influenza-like respiratory infections by boosting the immune system.
Electrochemical nitrogen reduction reaction (NRR) is a promising approach to convert earth‐adundant N2 into highly value‐added NH3. Herein, it is demonstrated that the heterogeneous Au–Fe3O4 nanoparticles (NPs) can be adopted as highly efficient catalysts for NRR. Due to the synergistic effect of the strong N2 fixation ability of Fe3O4 and the charge transfer capability of Au, the Au–Fe3O4 NPs show excellent performance with a high yield (NH3: 21.42 µg mgcat−1 h−1) and a favorable faradaic efficiency (NH3: 10.54%) at −0.2 V (vs reversible hydrogen electrode), both of which are much better than those of the Au NPs, Fe3O4 NPs, as well as core@shell Au@Fe3O4 NPs. It also exhibits good stability with largely maintained performance after six cycles. The N2 temperature‐programmed desorption, surface valance band spectra, and X‐ray photoelectron spectroscopy collectively confirm that Au–Fe3O4 NPs have a strong adsorption capacity for the reaction species and suitable surface structure for electronic transfer. The theoretical calculations reveal that Fe provides the active site to fix N2 into *N2H while introducing Au optimizes the adsorption of NRR intermediates, making the NRR pathway on Au–Fe3O4 along an energetic‐favorable process and enhancing the NRR.
The adsorption properties of coexistent Cr(VI) and Cu(II) in mixed solution on magnetic magnetite (Fe3O4) nanoparticle were studied in batch experiments. The influences of various factors, such as pH, adsorbent dose, temperature, initial concentration of metal ions, and coexisting ions in water were investigated. At the same time, the adsorption kinetics and adsorption isotherms were studied. The mechanism of adsorption for Cr(VI) and Cu(II) was investigated through the study of surface properties of Fe3O4, the presence of ions, and the influence of pH and zeta potential. The results indicate that pH has an influence on adsorption for Cr(VI) and Cu(II), and the optimal pH value for Cr(VI) and Cu(II) adsorption is 4.0. The adsorption efficiency increased with the increase of the adsorbent dose. Temperature under experimental design had no obvious effect. With 2.0 g/dm3 Fe3O4, the maximum adsorption capacity for Cr(VI) and Cu(II) reached 8.67 mg/g and 18.61 mg/g in mixed solution of 80 mg/dm3 Cr(VI) and Cu(II), respectively. Phosphorus had some influence on the adsorption for Cr(VI), and other coexisting anions and cations had no influence on the adsorption for Cr(VI) and Cu(II). The adsorption data for Cr(VI) and Cu(II) were nicely fit to the Langmuir adsorption equation and the pseudo-second-order model. As a multifunctional material, nano-Fe3O4 exhibited good adsorption performance for coexistent Cr(VI) and Cu(II) and could easily be separated and recovered under magnetic field.
