ВЯЧЕСЛАВ КУПРИЯНОВ К РУССКОЙ ФИЛОСОФИИ ЯЗЫКА (несокращенный вариант)/о работах академика РАО Ю. В. Рождественского/ Положение русского языка сегодня отражает не только падение интереса и уважения к России во всем мире, но и отношение самих русских к самим себе. Бывшие советские республики изгоняют русский язык из преподавания, надеясь таким образом избавиться от остатков его носителей, и таким образом не признают за русским языком культурообразующей роли. Ранее русский язык внедрялся как обязательный, что после "освобождения" от социализма стало восприниматься как насилие. Русский язык был объявлен передатчиком социалистической идеологии и не более, затем стал возбуждаться страх перед русским языком как воплощением русского империализма и шовинизма. В странах традиционно относимых к "западному миру", противостоящему России /вначале советской, а затем неважно какой/, все менее востребованной становится русистика, которая прежде была необходимым источником кадров как для открытой идеологической борьбы, так и подрывной деятельности. Податливое скатывание самой России в сторону сырьевой колонии сделало ненадобным переориентацию славистики на подготовку кадров в области сотрудничества на уровне новых технологий, а разбойничья репутация "новых русских" ослабила этот интерес даже в области торговли. Безработным славистам не осталось ничего более путного, как искать причину мытарств России в ее неправильной /недостаточно "западной"/ истории и культуре, ориентированной на мало кому внятное "византийство".Сам русский язык тем временем стал стремительно обогащаться ненормативной лексикой, о чем говорят тиражи словарей воровского жаргона и мата, который, хотя и был общеизвестен, но недостаточно продвинут в культурный речевой обиход благодаря исполнению советской милицией своего долга. А в знаменитом Тюбингенском университете я узнал от преподавателей, что в ученых кругах встал вопрос о введении русского мата в курсы русского языка и литературы. Что же касается так называемой русской интеллигенции, то она всегда старалась быть ближе к народу, чему мешала цензура, упразднение которой старанием перестройки можно считать фактом свершившегося единения интеллигенции с таким образом ею понимаемым народом.В этом смысле интересны молниеносно забытые призывы бывшего первого президента к интеллигенции: найти наконец и выразить в языке русскую идею. Кроме некоторых юмористов мало кто откликнулся на этот запоздалый призыв.Русская религиозная философия, выдвинувшая эту идею еще в XIX веке и старавшаяся ее развить в первой трети XX века, сегодня, наконец, вернулась на родину, ее изучают и пытаются толковать, для некоторых она последнее слово в русской мысли, при чем опускается факт, что эта мысль отражает состояние умов дореволюционных, а тем более -домассово-коммуникационных.Наша домашняя славистика скорее всего далека от всякой философии. The article is directed at developing and substantiating one of the approaches to forecasting the individual residual resource of a particular mining equipment in operation in a coal mine clean-up area. The main causes, affecting the accuracy of forecasts, have been identified. The perspectivity of using complex forecasts on several analogs with the help of spline functions for variables, characterizing the technological situations with GGOs, has been shown. The procedure of short-term forecasting of the multidimensional process of changing the average annual values of variables describing technological situations with the conveyor and lava supports in the breakage face has been adduced. The aim of the study was to investigate immunogenic properties of mosaic recombinant proteins constructed on the data of hepatitis C virus NS4A and NS4B antigens. Four mosaic recombinant proteins, containing the T and B epitopes of the NS4A and NS4B antigens, were created by genetic engineering methods in the E. coli system. To enhance the immune response they were linked in different variations to the nucleotide sequences of murine interleukin-2 (IL-2), the Neisseria meningiditis lipopeptide, and the T helper epitope of the core protein of hepatitis C virus. The immunogenic properties of these recombinant proteins were analyzed by immunoblotting, ELISA and ELISpot using sera from immunized mice and patients infected with hepatitis C virus. Recombinant proteins specifically reacted with the sera of immunized mice and infected patients in immunoblotting. According to the ELISA data, the predominant formation of antibodies to NS4B was observed when mice were immunized with the recombinant proteins containing both antigens. Analysis of gamma-interferon production by T-lymphocytes upon contact with activated dendritic cells showed in ELISpot that the maximum production of this cytokine was detected when adjuvant components were located at the N- and C-ends of the recombinant protein. The highest level of gamma-interferon production during stimulation with this drug was detected in lymphocytes from the bone marrow and lymph nodes. The recombinant protein containing the T and B epitopes of NS4A and NS4B, murine IL-2 and the lipopeptide Neisseria meningiditis had the greatest immunostimulate effect among the four constructions. This recombinant protein formed nanoparticles of 100-120 nm in size.
The article is directed to the development of one of the approaches to decision-making by the personnel on the meaningful analysis and interpretation of emergency situations in mine workings. The technique of quantitative estimation of non- stationary processes for detection of abnormal gas situations and emergency gassing has been offered. It has been focused on the full use of information redundancy, contained in the experimental implementations of the results of the interaction of technologies with the bottomhole part of the mountain massif. The following provisions were applied: the appearance of abnormal gas situations leads to a rupture of the interaction of gas dynamic processes at the sites of recording methanegrams, and the presence of a link indicates a «regular» development of situations. It has been shown, that, on the whole, the methodology is implemented in two stages: the evaluation of the significance of the connection by verifying the significance of the Spearman rank correlation coefficient when it is approximated by Student’s distribution and checking the stationarity of the observed connection at two time intervals of observation. It has been established, that the proposed technique, with appropriate modification, is also applicable to identification of soullars in mine workings. Two methods for monitoring gas evolution have been proposed. Verification of their content has been confirmed by mine experiments.
The aim of this study was to design promising variants of recombinant proteins based on NS4A antigen of hepatitis C virus (HCV) for subsequent work on the creation of a mosaic recombinant vaccine against hepatitis C. Methods. The recombinant proteins, containing different fragments of NS4A (belong to HCV subtype 1b) and murine interleukin-2, were prepared by genetic engineering approaches, using vectors pQE30 and pQE60 for E. coli. The size of the recombinant protein particles were evaluated by atomic force microscopy. Immunogenicity of these recombinant proteins was tested for Balb/c mice. The murine sera were analyzed by enzyme immunoassay. The recombinant proteins were also tested by immunoblotting with human sera specific to HCV antigens. Results. Six variants of recombinant genetic engineering constructions based on NS4A antigen of hepatitis C virus were designed. In the first variant amino acid sequence of NS4A was inserted using vector pQE60 into the immunodominant loop of HBc protein (core protein of hepatitis B virus). However, further analysis of the product showed the absence of virus-like particles in it. The following three constructs (with glycine linker 19s), without it and N-truncated NS4A) were done using vector pQE30. Only N-truncated NS4а product had a high expression level. Then new protein, consisted of NS4A and N-truncated murine interleukin-2 (IL-2), was obtained to enhance immunogenicity. It is known that IL-2 has adjuvant property. The new product (NS4a-IL-2) is well expressed, but it is accumulated in inclusion bodies. It was extracted with 7M guanidine chloride, purified on a Ni-sorbent and dialyzed in PBS. A shortened version of NS4A (ANS4a-IL-2) was also obtained with a high expression level. Taking in account that increasing the repetition of antigenic regions in recombinant constructs can enhance their immunogenicity, we obtained a recombinant protein comprising three repeat of NS4A. But its efficiency of expression was low. The construction NS4a had very poor immunogenicity, but NS4a-IL-2 (which contains the full length NS4A) displayed the best one for Balb/c mice. As it was shown earlier the immunogenicity of the protein preparation is dependent on the presence of aggregates, so we investigated our recombinant proteins for the presence of protein aggregates by atomic force microscopy.The presence of the particles with size of 6 - 8 nm was revealed in solution of NS4a-IL-2. Conclution. Only ANS4a-IL-2 and ANS4a-IL-2 of the six constructs had high expression and antigenic properties. And only NS4a-IL-2 possessed the high immunogenic property. So, this construction can be used for subsequent work on the creation of a mosaic recombinant vaccine against hepatitis C.
