Проанализировано экологическое состояние экосистемы озера Сабындыколь, дана гидрологическая и гидрохимическая характеристика водоема. Выявлено биологическое разнообразие фитопланктона и фитобентос а, зоопланктона и зообентоса, ихтиофауны, прибрежно-водной растительности. Рассмотрены донные отложения экосистемы озера. Отмечены усилившиеся в последнее время процессы заиления и обмеления водоема. Сток паводковых и ливневых вод со стороны расположенного рядом поселка приводит к загрязнению воды. Выпас скота по берегам озера способствует нарушению и уничтожению прибрежной растительности. Даны практические рекомендации природоохранного характера.
We performed a bioassay of aqueous extracts from the bottom sediments of the Chumysh River near the settlement of Talmenka (the Altai Territory of Russia) using the Allium test. The presence of toxic components that caused a decrease in the mitotic activity of onion root meristem cells was revealed. The frequency of pathological mitoses, significantly exceeding the background value (on average, 16.3 times), indicates the presence of factors with total mutagenic activity in the samples. We found violations of chromosome divergences (emissions beyond the spindle, lagging, running off and premature chromosome divergence), abnormalities of the mitotic apparatus (multipolar, asymmetric, monocentric mitosis, polyploidy), bridges (single and multiple). The prevalence of spindle pathologies suggests chemical contamination of the bottom sediments. Stress factors present in bottom sediments activate adaptive mechanisms of the test-object cells, aimed at maintaining their viability and reliability of the transmission of genetic information to the next cellular generations. Such adaptation mechanisms include the formation of micronuclei and cell polyploidization.
One of the modern methods widely used in the propagation of various garden crops is clonal micropropagation. Research in this direction is ongoing to optimize each stage of reproduction. The paper presents the results of a study of the clonal micropropagation of blackberry varieties Brzezina of Polish selection. For clonal micropropagation, sterile BlackBerry microshoots were used. In order to induce adventitious and axillary shoot formation, we tested various variants of culture medium: hormone-free medium, mediumsa with growth regulators (6-benzylaminopurine, β-indolyl-3-butyric acid, gibberellic acid). It was revealed that blackberry microshoots can take root on medium without growth regulators, but the resulting roots are thin, do not have root hairs. Such microshoots does not adapt to ex vitro conditions. A high reproduction rate (96%) was observed when rooting blackberry microshoots in a culture medium ½ MS + 2.0 μM IBA. Adaptation of regenerated plants to ex vitro conditions is more effective (97% survival rate) on a sand + vermiculite substrate (3: 1).
