We examined postoperative material from 28 patients aged 39-61 years with gliomas of different degrees of anaplasia (the diagnosis was histologically verified according to the WHO classification of CNS tumors) who had not previously received antitumor treatment. In glioma tissue, the glucose concentration was significantly higher than in the brain tissue of subjects dead from traumas (control), while lactate concentration did not differ from that in the control group or was lower. Hexokinase activity demonstrated a tendency to an increase in grade I and significant elevation in grades II and III, while in grade IV gliomas, this parameter did not differ from the control. Activities of the pentose-phosphate pathway enzymes glucose-6-phosphate dehydrogenase and transketolase increased with increasing of tumor anaplasia. Activity of glycogen synthase 3β kinase was significantly higher than in the control group. IDH1 mutation was discovered in 40% cases, the MGMT promoter methylation was detected in more than 50%, the Ki-67 level increased with increasing tumor anaplasia. The most significant correlations with glioma markers were detected for glucose-6-phosphate dehydrogenase and glycogen synthase 3β kinase. Activities of the studied enzymes of carbohydrate metabolism significantly correlated with Ki-67 marker.
This research was aimed at investigating the features of free radical activity and the parameters of glutathione metabolism in tumor tissues and the peritumoral zone at different degrees of glial tumor anaplasia. We analyzed postoperative material from 20 patients with gliomas of different degrees of anaplasia. The greatest differences compared to adjacent noncancerous tissues were found in the tumor tissue: an increased amount of glutathione and glutathione-related enzymes at Grades I and II, and a decrease of these parameters at Grades III and IV. For the peritumoral zone of Grades I and II, the indices changed in different directions, while for Grades III and IV, they occurred synchronously with the tumor tissue changes. For Low Grade and High Grade gliomas, opposite trends were revealed regarding changes in the level of glutathione and the enzymes involved in its metabolism and in the free radical activity in the peritumoral zone. The content of glutathione and the enzymes involved in its metabolism decreased with the increasing degree of glioma anaplasia. In contrast, free radical activity increased. The glutathione system is an active participant in the antioxidant defense of the body and can be used to characterize the cell condition of gliomas at different stages of tumor development.

Aim. Analysis of the electrophoretic mobility of red blood cells in patients with the diseases caused by increased cell proliferation activity, and its relationship with the lipid spectrum of erythrocyte membranes. Methods. The blood of 42 patients with malignant tumors of epithelial tissues (colon, kidney, prostate, urine bladder and cervical cancer) and 16 patients with skin diseases was studied. The control group consisted of 20 healthy individuals. Electrophoretic mobility of red blood cells was assessed by microelectrophoresis. Lipid fractions levels in erythrocytes was determined by one-dimensional thin layer chromatography. Results. Over than two-fold decrease of electrophoretic mobility of erythrocytes was observed in patients with malignant tumors of epithelial tissues and skin diseases associated with cell proliferation processes (psoriasis) - by 59 and 60%, respectively In inflammatory skin diseases (microbial eczema), the decrease was less pronounced (30%). The contents of charged phospholipid in erythrocyte membranes in case of cancer and psoriasis has changed in a similar way: a significant reduction in sphingomyelin level in the absence of significant differences in phosphatidylethanolamine and phosphatidylcholine levels. Multidirectional statistically significant changes in the contents of lysophosphatidylcholine were revealed depending on the stage of cancer, their level was decreased two-fold in case of psoriasis. The changes of cholesterol, diacylglycerols, free fatty acids levels in the membranes of red blood cells in case of cancer and psoriasis were divergent. Conclusion. In diseases caused by cell proliferation activation, electrophoretic mobility of erythrocytes is reduced by more than 50%. Kinetic characteristic of erythrocytes may be used for detecting the conditions of pathologic cell proliferation, including malignant tumors, since they alter in cases of phospholipid membranes lipid spectrum disorders.
