The objective of this study was to investigate the beneficial effects of mulberry extract (MBE) on blood flow improvement. The SC50 value for the DPPH radical scavenging activity of MBE was 89.36±5.46 μg/mL. Analysis of the cellular toxicity of MBE on RAW 264.7 and HepG2 cells showed no toxicity under a concentration of 2,500 μg/mL. We found that MBE inhibited the enzyme activity of cyclooxygenase (COX)-2 as well as oxidation of human LDL. Western blotting analysis showed that MBE inhibited protein expression of COX-2 and 5-lipoxygenase in RAW 264.7 cells. In addition, MBE inhibited protein expression of intercellular adhesion molecule-1 and vascular cell adhesion molecule-1 in human umbilical vein endothelial cells. Furthermore, MBE reduced the serum levels of total cholesterol and C-reactive protein in a concentration-dependent manner. These results both in vitro and in vivo suggest that MBE can be employed for the improvement of blood flow. 서 론 오디는 상심자, 상심, 상실, 오심이라고도 불리며 뽕나무 과(Moraceae), 뽕나무속(Morus)에 속하는 뽕나무(Morus alba L.)의 열매로서 식품공전에서 주원료(열매, 잎, 어린가 지)로 식품에 사용 가능한 동식물로 분류된다. 오디는 cyanidin-3-glucoside와 cyanidin-3-rutinoside와 같은 anthocyanin 계통의 색소를 다량 가지고 있으며(1,2), citric acid, malic acid 및 succinic acid와 같은 유기산과 polyphenol 등이 다량 함유되어 있다(3-5). 현재까지 확인된 오 디의 생리활성 기능들에는 항당뇨(6), 혈관신생억제효능(7), 항산화(8) 효과 등이 있다. 혈액순환장애는 혈액이나 림프액의 순환에 장애가 생기 는 질병이며 원인으로는 당뇨, 고혈압, 이상지질혈증, 고연 령, 가족력, 흡연, 비만, 운동부족 및 스트레스 등이 있으며 (9,10), 관련된 질환으로는 고혈압, 협심증, 심근경색증, 관 상동맥경화증, 고지혈증, 뇌졸중 및 뇌출혈 등이 있다(11-14). 혈액순환장애를 유발하는 인자들 중에서 이상지질혈증은 total cholesterol, triglyceride 및 LDL-cholesterol이 정 상보다 혈중농도가 높은 경우나 HDL-cholesterol이 정상 보다 혈중농도가 낮은 경우이다(15). 또한 체내 cholesterol 합성을 조절하는 효소인 HMG-CoA 환원효소(3-hydroxy-3-methyl glutaryl-coenzyme A reductase, HMGR)의 활 성화(16), 혈관협착을 유발하는 세포 간 접착분자인 ICAM-1(inter cellular adhesion molecule-1) 및 VCAM-1(vascular cell adhesion molecule-1)의 생성 증가(17), 동맥경 화증에서 정상보다 높게 나타나는 염증지표인자인 혈중 C-reactive protein의 농도(18), 혈전형성에 관여하는 물질 인 thromboxane A2(19), 대식세포와 같은 염증세포의 활성 으로 동맥경화증의 진행 및 죽상판의 취약성에 관여하는 효 소인 COX-2(cycloxygenase-2)(20) 및 혈전의 형성과 확 대에 관련된 LDL(low density lipoprotein)의 산화(21) 등 이 혈액순환장애에 관련되어 있다. 따라서 본 연구에서는 오디의 HMGR 활성억제 및 human LDL의 산화억제 효과를 예비실험에서 확인하고 오디 추출 물의 직･간접적인 혈행개선 효과를 체계적으로 검증하기 위 하여 in vitro에서 항산화 활성, human LDL의 산화억제 및 COX-2의 효소 활성 저해, 마우스의 대식세포주인 RAW 264.7 세포와 인간 간세포주인 HepG2에 대한 세포안전성 측정, 인간제대혈관내피세포(human umbilical vein endothelial cell, HUVEC)에서의 ICAM-1과 VCAM-1의 세포 내 발현억제 및 RAW 264.7 세포에서의 COX-2 및 5-LO의
