1,25(OH)2D3 and VDR Signaling Pathways Regulate the Inhibition of Dectin-1 Caused by Cyclosporine A in Response to Aspergillus Fumigatus in Human Corneal Epithelial Cells

Abstract: BackgroundThe objective of this study is to observe whether cyclosporine A (CsA) inhibits the expression of dectin-1 in human corneal epithelial cells infected with Aspergillus fumigatus (A. fumigatus) and to investigate the molecular mechanisms of the inhibition.MethodsImmortalized human corneal epithelial cells (HCECs) were pretreated with 1,25(OH)2D3 and VDR inhibitor for 1 h, and then they were pretreated with CsA for 12h. After these pretreatments, the HCECs were stimulated with A. fumigatus and curdlan r… Show more

“…128 The relationship of TLR4 with DR-driven repair mechanisms has been investigated by Vespasiani-Gentilucci et al, 129 showing a close correlation between TLR4 expression on HPC and DR cells and the inflammatory cell infiltrate, and deposition of fibrosis in liver biopsies of 74 patients with NAFLD. It is particularly noteworthy that TLR4 signaling is modulated by the vitamin D/Vitamin D receptor (VDR) pathway, acting as a fundamental protective mechanism not only in the liver, but even in the oral epithelium, 130 cornea, 131 and pancreas. 132 Similarly, reduced serological levels of 25-hydroxy-vitamin D3 and reduced tissue expression of VDR in both hepatocytes and cholangiocytes have been reported in both NAFLD 133 or NASH.…”

“…No entanto, diferenças entre gêneros, grupos étnicos e subtipos potencialmente diferentes dessa doença tornam difícil decifrar o papel exato da vitD na mesma. Além disso, embora a suplementação de vitD tenha sido bem-sucedida na melhora dos sintomas em alguns estudos, isso não foi demonstrado de forma consistente (XIA et al, 2016).…”
