1α,25‐Dihydroxyvitamin D3 increases IGF binding protein‐5 expression in cultured osteoblasts

Abstract: (BSA) (Sigma) was treated with charcoal to remove fatty acids and Abstract Rat osteoblasts produce insulin-like growth factors steroid hormones. Guinea pig anti-serum raised against human (IGFs) and IGF binding proteins (IGFBPs). Expression of IGFBP-5 was a generous gift from D.R. Clemmons, University of IGFBP-5, an IGFBP which stimulates DNA synthesis of North Carolina (Chapel Hill, NC) [13]. osteoblasts, was studied in vitro under the influence of hz,25-dihydroxyvitamin D3 (1,25(OH)2D3) and parathyroid hormo… Show more

“…Consistent with my findings for IGF-I, 1,25(OH) 2 D 3 was shown to mediate its stimulatory effect on osteoblast formation by increasing the production of IGF-I by human osteoblast cells [31] and mouse calvarial cells [32]. In addition, 1,25(OH) 2 D 3 upregulates many members of the IGFBPs including IGFBP-2, -3 and 4 in hMSC and IGFBP-5 in mouse osteoblastic cells [11,33]. On the other hand, the stimulatory effect of 1, 25(OH) 2 D 3 on IGFBP-6 is unique for NIH3T3 and has not been reported before for any osteoblastic cell line, and therefore might need further investigation.…”

“…Western ligand blot analyses revealed consistently the stimulation of secretion of the IGFBPs (182). 1␣,25(OH) 2 D 3 stimulates IGFBP-5 mRNA expression in cultured neonatal calvarial cells (198) and the osteoblastic UMR-106 cell line (199). Increased mRNA expression and secretion of IGFBP-3 and 4 have also been observed in human osteoblastic cells in several other studies (200 -203).…”

“…Several hormones, including prostaglandin E 2 and parathyroid hormone, enhanced IGFBP-5 gene expression in cultured osteoblasts [34][35][36], while transforming growth factor-b, platelet-derived growth factor, and basic fibroblast growth factor each reduced IGFBP-5 mRNA abundance in the same cells [37]. IGF-I also has been shown to induce IGFBP-5 mRNA and protein synthesis in cultured osteoblasts [19,33,34,37] and in bone in transgenic mice over-expressing IGF-I in osteoblasts [38].…”
