A (1→4)-β-mannan-specific monoclonal antibody and its use in the immunocytochemical location of galactomannans

Abstract: Galactomannan was coupled to a protein carrier for the preparation of monoclonal antibodies. The monoclonal antibodies generated bound to galactomannans from different sources as well as to glucomannan and galactoglucomannan. One monoclonal antibody, BGM C6, was characterised and found to be specific for (1-->4)-beta-linked mannopyranosyl residues; it had a binding affinity estimated at 1x10(-6) M for the (1-->4)-beta-linked mannohexaose. BGM C6 was used in immunogold labelling studies to locate galactomannans… Show more

“…In the case of the latter polysaccharides, an immunocytochemical approach to their detection with polysaccharide-specific antibodies has provided definitive proof of their synthesis in the Golgi (pectin and xyloglucan [Zhang and Staehelin, 1992]; mannan [Pettolino et al, 2001;Wilson et al, 2006]; AX [Philippe et al, 2006;Wilson et al, 2006]). This same approach has also been used to locate the site of MLG biosynthesis but has drawn contrasting conclusions in different Poaceae species.…”

“…Antibodies to MLG (Meikle et al, 1994), heteromannan (Pettolino et al, 2001), and (1,3)-b-D-glucan (Meikle et al, 1991) were from Biosupplies. A gold-conjugated goat anti-rabbit IgG secondary antibody (10 or 18 nm; Jackson ImmunoResearch) was used at 1:20 for visualization.…”

“…Grain samples from 10 to 28 DAP were prepared for microscopy and changes to the endosperm were monitored by taking transverse sections of the developing grains. The distribution and relative abundance of polysaccharides in the differentiating grain were detected on sections using monoclonal antibodies raised against specific polysaccharide epitopes including (1→3)-b-D-glucan (Meikle et al, 1991), xyloglucan LM15 (Marcus et al, 2008), (1→3,1→4)-b-D-glucan (Meikle et al, 1994), hetero-(1→4)-b-Dmannan (Pettolino et al, 2001), and arabino-(1-4)-b-D-xylan LM11 (McCartney et al, 2005). Sections for light microscopy (1 mm) were either stained in toluidine blue to highlight tissue architecture or processed for immunogold labeling.…”

“…In our previous work, LM10 and LM11 antibodies showed some differences in AGX localization (Kim et al, 2010c). Some sections were also incubated with BGM C6 monoclonal antibody specific to (1→4)-β-linked mannopyranosyl residues (Pettolino et al, 2001). Thereafter, grids were incubated with goat anti-rat (LM10 and LM11) or anti-mouse (BGM C6) secondary antibody labeled with 10-nm colloidal gold particles for 1 h at room temperature.…”
