A 28 nt long synthetic 5′UTR (synJ) as an enhancer of transgene expression in dicotyledonous plants

Abstract: BackgroundA high level of transgene expression is required, in several applications of transgenic technology. While use of strong promoters has been the main focus in such instances, 5′UTRs have also been shown to enhance transgene expression. Here, we present a 28 nt long synthetic 5′UTR (synJ), which enhances gene expression in tobacco and cotton.ResultsThe influence of synJ on transgene expression was studied in callus cultures of cotton and different tissues of transgenic tobacco plants. The study was base… Show more

“…and the exact position of the structural domain needs to be verified by additional experiments. Previous studies reported that 5´-UTR could act as an enhancer in the gene expression process (Viana et al 2011;Kamo et al 2012;Kanoria and Burma 2012;Agarwal et al 2014). In absence of 5´-UTR region, MH (-1057 to -266) had a weaker GUS activity than MH10, which implies that 5´-UTR may regulate the MdHB-1 gene expression.…”

“…From these, two datasets were developed comprising of 600 bp Upstream Regulatory Module (URM), one from dicot and another from monocot species. URMs [40] are defined as a region of a gene upstream to the translational start site, which includes the 5'UTR. Analyzing the URM was necessary in this analysis as in most cases the transcriptional start site has not been experimentally identified.…”

“…However, it remains a mystery how the diversity of these sequences affects the efficiency of translation [7]. Currently, the 5'-UTRs and their influence on the efficiency of gene expression is being studied intensively [8,9]. Basically, comparative studies of 5'-UTRs of various genes are being conducted using reporters [10,11].…”
