A Bioanalytical Method for Eliglustat Quantification in Rat Plasma

Abstract: A simple and sensitive bioanalytical HPLC–UV method has been developed and validated for quantification of eliglustat in rat plasma. The liquid–liquid extraction method was found to be more efficient compared to protein precipitation technique. Chromatographic separation of eliglustat was achieved using Kromasil C18 column with a mobile phase consisting of a mixture of methanol and ammonium acetate (pH 3.2) in a ratio of 60:40. Detection wavelength was set at 282 nm. The developed method was specific, accurate… Show more

“…In the present study, we developed and established a UPLC-MS/MS method for the determination of eliglustat in rat plasma. To the best of our knowledge, only one paper had described a bioanalytical method of determining eliglustat in plasma by HPLC prior, which required elaborate sample preparation (liquid–liquid extraction), low sensitivity (300 ng/mL), and long run time (12 minutes) 18. We instead chose a simple protein-precipitation method using acetonitrile.…”

“…Three chemical structures of ELGS stereoisomers, namely, (1R,2S)-stereoisomer, (1S,2R)-stereoisomer, and (1S,2S)stereoisomer, are represented in Figure 1. [3,4] From the review of literature, there are only two highperformance liquid chromatography (HPLC) methods [5,6] that have been reported for the estimation of ELGS and no chiral HPLC method iswas reported for the quantitation of ELGS and its stereoisomers. In the present research paper, chromatographic method was developed with immobilized Amylose tris (3-chloro-4-methylphenylcarbamate) chiral stationary phase namely CHIRALPAK IF-3(4.6 mm × 250 mm, 3 µm), utilized for the separation and quantification of ELGS isomers.…”

“…The symptoms and progression of RA can be controlled by treatment strategies, which include non-steroidal anti-inflammatory drugs (relives from pain and stiffness), corticosteroids (minimize the inflammatory events) and DiseaseModifying Antirheumatic Drugs (DMARDs) (suppress progression of RA). 24 However, rheumatologists preferred to prescribe combination therapy rather than monotherapy because of the increased efficacy and enhanced improvement of the patient over time. 5 Dell et al performed a two years study and measured the difference in the improvement of patients with three different treatments i.e, Methotrexate (MXT) alone, Sulfasalazine (SFS) and Hydroxychloroquine (HCQ) combined treatment and combination of all three.…”
