A Bufadienolide-Enriched Fraction of Bryophyllum pinnatum Inhibits Human Myometrial Contractility In Vitro

Abstract: Bryophyllum pinnatum has been used since the 1970s to prevent premature labour, first in anthroposophic hospitals and, more recently, also in the main Swiss perinatal centres. However, it is not known which compounds in B. pinnatum leaves contribute to the tocolytic effect. Here we studied the effects of a flavonoid-enriched fraction, the corresponding flavonoid aglycon mixture, a bufadienolide-enriched fraction, and B. pinnatum leaf press juice on human myometrial contractility in vitro. The strength (area un… Show more

“…The MPFF effects in the short term were as expected in all experimental groups. Accordingly, the anti-inflammatory effects of other FGs decreased within weeks of administration, as reported for several species such as murine and humans [ 59 , 60 ].…”

“…2, 3 and 4). Our previous work suggests that various components of B. pinnatum leaves might contribute to increase contraction frequency of myometrium strips [24]. Both a bufadienolide-enriched fraction and a flavonoid-enriched fraction (but not the corresponding flavonoid aglycon mixture) seemed to contribute to the frequency increase.…”

“…To find out whether the effect of the combination treatments would further increase at higher concentrations, a previously described approach was followed [24]. In brief, when spontaneous contractions were regular for 20 min, Krebs solution was added (addition 0), and contractility was recorded for 20 min.…”

“…B. pinnatum is known to act on uterine contractility. In vitro research with human myometrial cells has been conducted at university centers in Switzerland since the early 2000s [3,29,30,55,56]. Several clinical studies have also evaluated the tocolytic effect of B. pinnatum in premature labor [44,45,57].…”
