A Combinatorial Approach to Catalytic Peptide Dendrimers

Abstract: Exploring the structural diversity of peptide dendrimers as synthetic protein models: A 65 536‐membered combinatorial peptide‐dendrimer library was prepared by split‐and‐mix techniques on beads (see picture). The library was screened and revealed peptide dendrimers that catalyze fluorogenic ester hydrolysis and peptide dendrimers that bind to vitamin B12.

“…5 Split-and-mix synthesis makes possible the generation of a large number of compounds with only a few synthetic steps. 6 The method generates single beads displaying only one type of compound and can be applied for the screening of millions of different compounds while still attached to the beads.…”

“…An efficient process of this kind has been reported using an organic polymer consisting of modified polyethyleneimine [209], with important deductions on the capabilities of primitive enzyme catalysis. A noticeable efficiency and selectivity has been reported for selected reactions of peptide dendrimers [210,211], in which the sequence and, possibly, the structure are more precisely defined. These kinds of catalytic effects may be related to the reactions that could have been promoted by uncoded prebiotic peptides.…”

“…[68][69][70] Dendrimers can also act like protein, with their internal cavities and surface channels. [71][72][73][74][75][76] As an exo host molecule, the guest interacts with the functional group located at the surface of the dendrimers like metal ion complexations. The dendrimers can be used as a complex and large ligand for catalytic species.…”
