A comparative study of chiral separation of proton pump inhibitors by supercritical fluid chromatography and high‐performance liquid chromatography

Abstract: A comparative study of chiral separation of pantoprazole and rabeprazole is carried out using supercritical fluid chromatography and high‐performance liquid chromatography. The columns used were Chiralpak IA and Chiralpak IE. The best mobile phase in supercritical fluid chromatography was carbon dioxide‐0.2% triethylamine in methanol (60:40) and 0.1% triethylamine in n‐hexane‐ethanol (50:50) in high‐performance liquid chromatography. For supercritical fluid chromatography, values of the retention factor of pan… Show more

“…Among many chiral stationary phases (CSPs), polysaccharide-based CSPs are the best owing to their remarkable recognition powers. [25][26][27][28] During the search for analytical techniques for this purpose, supercritical fluid chromatography (SFC) was considered the best as a consequence of its ease of operation and inexpensive nature. Furthermore, the purity of the peaks was ascertained by Chiral-liquid chromatographymass spectrometry (LC-MS).…”

“…These chiral columns such as Chiralpak IA and Chiralpak IG are amylose-based chemistry resulting in efficient enantiomeric separation. 7,8 Therefore, some potential antibiotic drugs such as ofloxacin and cefotaxime were selected for the chiral separation using aforesaid chiral stationary phases (CSPs). Ofloxacin belongs to the fluoroquinolones group and is also known as a synthetic anti-microbial agent (Figure 1).…”
