The distribution of soil ciliates in three different habitats within a typical mangrove forest in Dongzhaigang, Hainan, China was investigated. The abundance, biodiversity, and community similarity of ciliates in fresh and air-dried soil with different, physical/chemical properties were analyzed. Three Classes, 11 Orders, 34 Genera, and 70 species of ciliates were found with the first dominant group being Hypotrichida. Ciliate biodiversities followed Site B < Site A < Site C in both fresh and dried samples. Ciliate abundance was positively correlated with soil moisture, salinity, organic matter (OM), total nitrogen (TN), total phosphorus (TP), and sulfate (SO 4 2− ), but negatively with pH and total potassium (TK). Site A and Site B and Site B and Site C showed the highest similarity in fresh and dried samples, respectively. The ubiquitous characteristics of ciliate distribution suggested their important role in food webs and nutrient cycling. The presence of Colpodida was linked with mangrove plants.

Biological soil crusts are diverse assemblages of bacteria, cyanobacteria, algae, fungi, lichens, and mosses that cover much of arid land soils. The objective of this study was to quantify protozoa associated with biological soil crusts and test the response of protozoa to increased temperature and precipitation as is predicted by some global climate models. Protozoa were more abundant when associated with cyanobacteria/lichen crusts than with cyanobacteria crusts alone. Amoebae, flagellates, and ciliates originating from the Colorado Plateau desert (cool desert, primarily winter precipitation) declined 50-, 10-, and 100-fold, respectively, when moved in field mesocosms to the Chihuahuan Desert (hot desert, primarily summer rain). However, this was not observed in protozoa collected from the Chihuahuan Desert and moved to the Sonoran desert (hot desert, also summer rain, but warmer than Chihuahuan Desert). Protozoa in culture began to encyst at 37 degrees C. Cysts survived the upper end of daily temperatures (37-55 degrees C), and could be stimulated to excyst if temperatures were reduced to 15 degrees C or lower. Results from this study suggest that cool desert protozoa are influenced negatively by increased summer precipitation during excessive summer temperatures, and that desert protozoa may be adapted to a specific desert's temperature and precipitation regime.

Techniques for examining pore structure as a three-component system, most notably dyestaining of pore space, are reviewed. A method for analysing digital images of pore structure with three components is presented. The method is a direct extension of earlier work on the description of pore structure using two-component digital images in which the two components represented pore space and soil solid. A previously published method for preparation of an undisturbed, vertically oriented, planar face through soil with pore space in the field condition is used. Field impregnation with opaque epoxy resin is followed by sawing, completion of the impregnation under vacuum and grinding of the sawn face to a smooth finish. In the method presented here the resin used for the laboratory impregnation contains a different-coloured dye from that used in the field. The ground face is then video-digitized. Three components are represented in the resulting digital grey level image, namely, field-impregnated pore space, laboratory-impregnated pore space and soil solid. The image-processing steps required to segment the grey level image into the three required components are described. Image analysis, for quantitative assessment of the pore structure, consists of generation of 1 1 pore-structure attributes from the segmented image. Interpretation of these attributes and use of the method are discussed.
