A differential stain for the anterior lobe of the hypophysis

Abstract: A considerable number of investigations have been published which have demonstrated that the anterior lobe of the hypophysis plays an important rBle in the physiology of the female reproductive system. Some of these studies have shown not only that the hypophysis is an important part of

“…In addition, immunostaining was abolished when the antisera were omitted or preabsorbed with the corresponding antigens, while cross-absorption of the antisera with the non-corresponding peptide had no effect. For histological and histochemical study, the sections were stained with: (1) Herlant's alcian blue-periodic acid-schiff (PAS)-orange G (Herlant II;Herlant, 1960); (2) Gomori's chromium-haematoxylin-phloxin (CHP) (Gomori, 1941); (3) Cleveland-Wolfe trichrome (CW) (Cleveland & Wolfe, 1932); (4) Azan-Heidenhein (Azan) (Koneff, 1938); (5) lead haematoxylin (PbH) (Solcia et al, 1969) with or without PAS or (6) PAS-haematoxylin (PASH).…”

“…All pituitaries were fixed by the method of Severinghaus (1932), of Cleveland and Wolf e (1932), and were stained after the same authors' methods. Paraffin sections were cut serially at 3,~ in the sagittal plane.…”

“…Mercury was removed from the tissues fixed in the Bouin-Hollande fixative by immersing the slides in iodine (1%) in 70% alcohol, followed by sodium thiosulfate (about 5%) and then rinsing in tap water and distilled water. In order to visualize the thyroid gland development, the sectioned larvae were stained using Cleveland-Wolfe trichrome staining (Cleveland and Wolfe 1932). The tissue sections were stained in Erlich's haematoxylin for 5 min, then ''blued'' by a short immersion in alkaline alcohol to stain nuclei, rinsed in tap water and distilled water, followed by staining in 1% aqueous erythrosine for 5 min.…”
