A direct organogenesis protocol from shoot segments of Solanum tuberosum cv. Monalisa

Abstract: Transgenic technology is an excellent alternative for improvement of crop production and disease free crops such as potato, which is one of the most important crops worldwide. One of the first steps to apply the transgenic technique is the establishment of an efficient plant regeneration protocol. This is a limiting step in this kind of study, since most protocols are species-specific and some of them do not adequately respond to in vitro culture or present low regeneration rates. The objective of this study w… Show more

“…Hence, in the current study, we evaluated direct shoot regeneration from four potato genotypes of Kazakhstan breeding using two different explants. Our results are in agreement with studies demonstrating that GA 3 isan important plant hormone with the ability to induce a number of responses, including direct shoot regeneration ( Ibragimova et al, 2018 ; Roest & Bokelmann, 1976 ), and suggesting that the combination of auxins and cytokinins in the presence of GA 3 enhances shoot regeneration from explants ( Campos et al, 2016 ; Kumlay, 2014 ).…”

“…Many studies have reported that often most of the tissue culture process is cultivar-specific. It is, therefore, necessary to establish a specific protocol for each genotype studied ( Campos et al, 2016 ; Tara, Krishnaprasad & Veena, 2017 ; Yadav & Sticklen, 1995 ).…”

“…Shoot organogenesis protocol, a pre-requisite for genetic manipulation of plants has been well documented in potato but is complicated by use of two step procedure (Yee et al 2001, Yasmin et al 2003, Elaleem et al 2009, Molla et al 2011, Rezende et al 2013, Ghosh et al 2014, Abdelaleem 2015, Campos et al 2016. Moreover, genotype specific response of potato cultivars to plant growth regulators (PGRs) has also been reported by many researchers (Khokan et al 2009, Pal et al 2011, Wheeler et al 1985.…”

“…In potato, an important rabi food crop, shoot organogenesis is extensively studied using different explants such as leaf, internode and tuber discs (Yasmin et al 2003, Elaleem et al 2009, Molla et al 2011, Rezende et al 2013, Ghosh et al 2014, Abdelaleem 2015, Campos et al 2016, Kaur et al 2017). Most of the reports are mainly focussed on Plant Growth regulators (PGRs), but, studies on the role of other factors such as gelling agents, light intensity, medium pH, silver nitrate etc.…”
