A fatal affair: Circulating tumor cell relationships that shape metastasis

Abstract: Circulating tumor cells are metastatic precursors in several cancer types. Their biology and clinical utility are subject to numerous investigations, yet one aspect that is often neglected is their entanglement with the tumor microenvironment, namely the cross talk with stromal and immune cells and their relationships with other tumor-derived components such as circulating tumor DNA and extracellular vesicles in circulation. We will focus our short review specifically on these aspects, i.e., providing some exa… Show more

“…Single-cell DNA methylation sequencing has been widely used to profile rare cells and investigate cellular heterogeneity. CTCs preserve tumor genetic and epigenetic information well and are excellent candidates for cancer prognosis and diagnosis [138][139][140]. It is foreseeable that single-cell methylation sequencing and site-specific methylation assays will be incorporated into clinical testing.…”

“…Cluster formation is produced in part by contact between a cell-cell ligand-protein or cytokine receptor and another cell [74]. An aggregate of more than one cancer cell is a homotypic CTC cluster; in combination with other cell types, such as immune cells or stromal cells, are heterotypic CTC clusters [75]. Compared to the single cells present in the circulation, these CTC clusters have an intercellular adhesion component that can promote tumor metastasis [76].…”

“…A versatile and facile strategy for evaluating drug sensitivity of cancer patients is highly needed.Circulating tumor cells (CTCs) shed from solid tumor lesions and circulate in the bloodstream, and they contain the whole information relating to their origin. [9][10][11] Currently, CTCs can be enriched from vein blood non-invasively by various strategies [12][13][14] and are expected to reflect drug sensitivity of the tumors. [15,16] A pharmacogenomic profiling technology was developed to reveal gene expression of CTCs to identify pancreatic cancer patients who were sensitive to chemotherapy regimens.…”
