Background:Anti-inflammatory effect of natural adjuvants has been reported. Lung inflammation is the most characterized pathological feature in asthma.Objectives:The effects of three natural adjuvants (PC, G2, and G2F registered as a patent in the Iranian Patent Office) on sensitized guinea pigs lungs were examined in the present study.Materials and Methods:Lung pathological changes were examined in control and five groups of randomly divided guinea pigs including: sensitized animals (S, receiving normal saline, 0.5 ml i.p.); sensitized animals treated with adjuvant PC; G2F (0.1 ml i.p. for both cases); G2 (0.4 ml i.p.); and PC + G2 (receiving both PC and G) adjuvants (twice a week for 4 weeks for all groups). Sensitization of animals was done by injection and inhalation of ovalbumin (OA).Results:All pathological changes in S group including the eosinophil infiltration (scoring 3.28 ± 0.28), lymphocyte infiltration (2.82 ± 0.26), local epithelial necrosis (2.71 ± 0.47) and mucosal plug (2.75 ± 0.37) were significantly higher than control group (0.64 ± 0.18, 1.36 ± 0.24, 0.36 ± 0.18 and 0.28 ± 0.18 for eosinophil infiltration, lymphocyte infiltration, epithelial necrosis and mucosal plug respectively, P < 0.001 for all cases). Treatment with all adjuvants improved all pathological changes significantly (P < 0.05 to P < 0.001).Conclusions:These results indicate preventive effects of all natural adjuvants (especially G2) on pathological changes of the lung in sensitized guinea pigs.

The current study investigated the effects of phytochemicals genistein and/or hesperidin dietary supplementation on immunity and haematology of growing broilers. A total of 360 1-day-old broiler chicks (Arbor Acres, mixed sex) were randomly assigned to six treatment groups, namely T0, control; T1 and T2, supplemented with 5 and 20 mg of genistein and hesperidin; while T3, T4, and T5 diets contained 5, 10, and 20 mg of genistein þ hesperidin (1:4) mixture, respectively, per kg of diet. The white blood cell count was significantly (p < .01) increased in T1, T2, T4, and T5 compared with the control (T0) group. The haemoglobin concentration significantly (p < .01) increased in the T5 group, while mean corpuscular haemoglobin concentration was significantly (p < .05) higher in T4 compared with the T0 group. Antibody titres against Newcastle disease significantly (p < .01) increased in T1, T2, T3, T4, and T5 compared with the T0 group. Similarly, all phytochemicals treated groups exhibited an increase (p < .01) in antibody titres against Avian Influenza virus, as compared with the controls. At the same time, the supplemented groups had significantly (p < .01) higher neutrophil adhesion rate and cutaneous basophil hypersensitivity test representing the cellular immune response than the controls. In conclusion, supplementation with both phytochemicals, genistein and hesperidin, positively influenced the immune parameters and haematological profile of growing broilers, thus might be considered as feed additives in broiler industry. Genistein and hesperidin have been shown to have beneficial effects in several animal models including mice, rats, pigs, and humans. This study investigated the individual and combined effects of genistein (an isoflavone) and hesperidin (a flavanone) on immunity and intestinal morphometry in lipopolysaccharide (LPS)-challenged broiler chickens. Seven hundred twenty 1-d-old commercial Arbor Acres broiler chicks were randomly divided into 6 treatments, with 6 replicates of 20 birds each. Chicks were fed a basal diet without any additive (control), supplemented with 5 mg of genistein/kg of feed (G5) and 20 mg of hesperidin/kg of feed (H20), or a mixture of genistein and hesperidin (1:4) with doses of 5 (GH5), 10 (GH10), and 20 (GH20) mg/kg of feed for 42 d. On d 16, 18, and 20, one-half the birds from each group were separated and injected intraperitoneally with Escherichia coli LPS (250 µg/kg of BW) to induce the immunological stimulation. Samples were collected on 21 and 42 d. The results showed that LPS treatment exerts immunomodulatory effects (P < 0.05) in phagocytic activity at 21 d, whereas a few negative effects including reduced villus length and increased crypt depth were observed in some segments of the small intestine. Both genistein and hesperidin seemed to modify the immunity positively by altering the phagocytic activity (P < 0.01). Parameters of intestinal morphometry such as villus length, crypt depth, villus width, and villus length/crypt depth ratios were also improved (P < 0.01 or P < 0.05) by the supplemental genistein and hesperidin in both LPS-unchallenged and -challenged groups. However, no effect (P > 0.05) was observed for BWG, FI, and FCR of broilers. Overall, genistein and hesperidin improved the immunity and the morphometry of small intestine in a dose-dependent manner. These findings provided the first account on the in vivo effects of genistein and hesperidin for immunostimulation and morphometric gut development in LPS-challenged chickens. Thus, both compounds may be used as alternative feed additives in the poultry industry to promote gut health and improve immunity against infections.
