A fluorescence and UV/vis absorption dual-signaling probe with aggregation-induced emission characteristics for specific detection of cysteine

Abstract: A dual-signaling of fluorescence and UV/vis absorption modes for selective and quantitative detection of cysteine over homocysteine, N-acetyl-l-cysteine and glutathione is developed on the basis of aggregation-induced emission (AIE) effect.

“…To prepare TPR, TPE-COOH was first synthesized according to the reported procedures (Figure S1). 36,37 The characteristic peaks of H in different environments clearly emerged in the 1 H nuclear magnetic resonance (NMR) spectrum (Figure 2A), confirming the successful preparation of TPE-COOH. With the aid of the standard solid-phase synthesis method, TPR was elaborately prepared by sequential coupling amino acids, NH 2 -PEG 8 -COOH, and TPE-COOH.…”

“…According to the published literature, the possible reaction mechanism between MITO-PQDNs and GSH was presented in Scheme 1. 20,22,[37][38][39] The strong electron-absorbing DNs group signicantly quenched the uorescence of MITO-PQDNs. However, the strongly nucleophilic GSH could undergo a nucleophilic aromatic substitution reaction (SNAr) with MITO-PQDNs leading to the cleavage of the sulfonate esters, thus releasing the uorophore MITO-PQ.…”

“…When constructing S N Ar-based RSS- and RSeS-specific fluorescent probes, an ether linkage is a popular choice as the reaction site [251] , [252] , [253] , [254] , [255] , [256] , [257] , [258] , [259] . Phenyl-O ether, phenyl-S ether, and phenyl-Se ether have mainly been used, among which the C-O, C-S, and C-Se bonds become increasingly long [161] .…”
