A Genetic Tool Used to Identify Thioredoxin as a Mediator of a Growth Inhibitory Signal

Abstract: Loss of sensitivity to growth inhibitory polypeptides is likely to be one of the events that participates in the formation of some tumors and might be caused by inactivation or loss of the genetic elements that transduce these extracellular signals. The isolation of such a gene was achieved by randomly inactivating genes by an anti-sense complementary DNA expression library followed by direct selection for growth in the presence of an inhibitory polypeptide. Thus, a gene whose inactivation conveyed growth resi… Show more

“…In this strategy cells are transfected with a total antisense cDNA library cloned in an episomal vector and then selected with IFN-b/RA (Deiss and Kimchi, 1991;Kalvakolanu, 2004). As the antisense RNAs derived from this library inactivate the expression of endogenous death-associated mRNAs, cells become resistant to death stimulation by IFN-b/RA, and continue to proliferate.…”

“…Ligated to the 3Ј and 5Ј ends of the cDNA, this linker generates HindIII and BamHI sites, respectively (25). The library was digested with HindIII and BamHI, purified on a Sepharose 6B column, and ligated into an episomal vector pTKO1 (from Adi Kimchi, Weizmann Institute, Rehovot, Israel), which carried markers for selection in eukaryotic and bacterial cells (22). When cloned into pTKO1 the cDNA is expressed in antisense orientation.…”

“…To identify death genes we used an antisense technical knockout approach (22). In this approach, death regulatory genes are identified by their ability, when expressed in antisense orientation, to confer resistance to death inducers.…”
