A Genome-Wide RNAi Screen to Dissect Centriole Duplication and Centrosome Maturation in Drosophila

Abstract: Centrosomes comprise a pair of centrioles surrounded by an amorphous pericentriolar material (PCM). Here, we have performed a microscopy-based genome-wide RNA interference (RNAi) screen in Drosophila cells to identify proteins required for centriole duplication and mitotic PCM recruitment. We analysed 92% of the Drosophila genome (13,059 genes) and identified 32 genes involved in centrosome function. An extensive series of secondary screens classified these genes into four categories: (1) nine are required for… Show more

“…In Drosophila S2Rϩ cells, RNAi knockdown of Bld10 gave an intermediate centriole replication phenotype (Dobbelaere et al, 2008). This contrast with our in vivo results, likely reflects a peculiarity of S2Rϩ cells or possible off-target effects of RNAi.…”

“…These data indicate that Bld10 is likely a ubiquitous component of centrioles throughout development. Two recent reports corroborated the centriole localization of Bld10 (Blachon et al, 2008;Dobbelaere et al, 2008).…”

“…Many proteins are involved in the regulation of centrosome maturation 10 and, importantly, disruption of these proteins leads to defects in spindle formation similar to the defects seen with Kin-1 depletion [14][15][16] . Such proteins are collectively known as PCM proteins and include Plk-1, Cdk5rap2 and Aik-A.…”

“…In flies, where the pathway of centrosome maturation has been more extensively studied, the Pericentrin-like protein (D-PLP) shares this role with centrosomin (Cnn; Martinez-Campos et al 2004). The localisation of both these PCM proteins is dependent upon the presence of the core centriolar component D-Spd2 (Dix and Raff 2007;Giansanti et al 2008), whilst the phosphorylation of Cnn by the mitotic kinase, Polo, correlates with centrosome maturation (Dobbelaere et al 2008), suggesting a hierarchical pathway in which γ-tubulin is targeted and maintained at the PCM in a stepwise fashion.…”
