A genomic strategy for the functional validation of colorectal cancer genes identifies potential therapeutic targets

Abstract: Genes that are highly overexpressed in tumor cells can be required for tumor cell survival and have the potential to be selective therapeutic targets. In an attempt to identify such targets, we combined a functional genomics and a systems biology approach to assess the consequences of RNAi-mediated silencing of overexpressed genes that were selected from 140 gene expression profiles from colorectal cancers (CRCs) and matched normal mucosa. In order to identify credible models for in-depth functional analysis, … Show more

“…Transfections with synthetic small interfering RNA (siRNA) duplexes were performed as previously described (16,17). Briefly, for cellular viability assays, cells were reverse transfected with siRNAs (Qiagen, Hilden, Germany; Dharmacon/Thermo Fisher Scientific, Schwerte, Germany) using RNAiMAX (Invitrogen) or HiPerFect (Qiagen).…”

“…Overexpression of RRM2 leads to lung cancer in mice, and elevated RRM2 in ovarian, colorectal, liver, and breast cancers is associated with poor prognosis. [108][109][110][111][112] Downregulation of the cyclin G2 transcript has been linked to various cancers including the thyroid and oral cavity. 113,114 Cyclin G1 and cyclin G2 share 53% amino acid identity; however, cyclin G1 lacks the protein-destabilizing PEST domain.…”
