A Glucosamine-Specific Lectin from Green Dragon No. 8 Beans (Phaseolus vulgaris) Induced Apoptosis on Nasopharyngeal Carcinoma Cells

Abstract: A lectin exhibiting antiproliferative activity on tumor cell lines but devoid of antifungal activity has been purified from Phaseolus vulgaris cv. Green Dragon no. 8 seeds. The lectin was a 60 kDa dimeric protein with two 30 kDa subunits. It was a glucosamine-specific lectin as implied from the inhibitory effect of glucosamine on hemagglutinating activity of the lectin. The steps for isolation of the lectin involved Affi-gel blue gel (affinity gel), Mono Q (anion exchanger), and Superdex 75 column (size exclus… Show more

“…A lectin from Phaseolus vulgaris L. 'French bean number 35' induce apoptosis in MCF-7 cells [91]; and a polygalacturonic acid-specific lectin, named BTKL, isolated from seeds of Phaseolus vulgaris L. "Blue tiger king", exerted a potent antiproliferative action on Hep G2 cells [92]; and Phaseolus vulgaris L. 'Green Dragon number 8' beans contain a glucosamine-binding lectin that could inhibit nasopharyngeal carcinoma HONE1 cells after treatment for 24 h (IC50 = 62.3 µM), 48 h (IC50 = 31.6 µM), and 72 h (IC50 = 28.7 µM), preventing the cells from dividing and kept them at the G2/M phase. It also had an effect on nasopharyngeal carcinoma CNE2 cells, after 24 and 48 h treatment [93]. The EAPL showed antitumor activity against nasopharyngeal carcinoma (CNE-1, CNE-2, HNE-2) cell lines, breast cancer cells (MCF-7) e hepatocellular carcinoma (HepG2), inhibiting their proliferation in a dose dependent manner.…”
