A highly selective two‐stage isolation method for the enumeration of Staphylococcus aureus in foods

Abstract: A plate method for enumerating Staphylococcus aureus is described which combines a 1‐h recovery period for stressed cells on a relatively non‐selective Baird‐Parker agar base followed by a 24‐h growth phase in a highly selective, supplemented Baird‐Parker medium added as an overlay. Tests with pure cultures showed satisfactory recovery of stressed Staph. aureus and other bacteria. Similar results were obtained with the conventional Baird‐Parker procedure and with the two‐stage isolation method for shrimps and … Show more

“…The acriflavine MICs were in the range 0?8-1?6 mg ml 21 for the four novel isolates, whereas they amounted to 12?5 and 25 mg ml 21 for S. aureus strains (Devriese, 1981). S. aureus strains are unique in being the only staphylococci described, to date, that are resistant to more than 8 mg acriflavine ml 21 -a level used in the selective isolation of this species in food microbiology (Isigidi et al, 1989).…”

“…A second factor that influences the numbers of organisms recovered is whether a resuscitation treatment (Mossel & van Netten 1984;Ray 1989) is used. In many instances staphylococci are severely injured, so that recovery is erroneously low if a specimen is plated directly on many currently used selective media (Idziak & Mossel 1980;Mossel et al 1985;Isigidi et al 1989;van Netten et al 1990). Such cells have to be included in counts as when they have recovered from injury their enterotoxinogenicity is unimpaired (Fung & van den Bosch 1975;Smith et al 1984).…”
