A Less Invasive Approach of Medial Meniscectomy in Rat: A Model to Target Early or Less Severe Human Osteoarthritis

Abstract: Objectives: The existing medial meniscectomy (MMx) procedure in rodents involves transection of MCL and wide opening of the knee capsule followed by meniscus transection that results into articular cartilage degeneration and causes significant changes in subchondral bone at specific post-MMx period. These animals serve as experimental osteoarthritis (OA) models.Structurally, knee is very complex. There may be a need to uncouple articular cartilage degenerative changes from subchondral bone degenerative changes… Show more

“…Seven-week-old Slc:Wistar male rats were housed under a 12-h light cycle in a temperature-controlled room for 1 week before surgery was performed. Experimental OA was induced by MMx 28 . Briefly, anesthesia was induced in rats [medetomidine (2 mg/kg bodyweight), midazolam (0.4 mg/kg), butorphanol (5 mg/kg)].…”

“…Even though chitosan shares structural similarities with the glucosamine glycans (GAGs) present in PGs, which have been shown to stimulate chondrogenesis [ 34 ], chitosan seems to interact with growth factors to stimulate fibrocytes and no other cells [ 35 , 36 ]. This may explain why despite an increase in the viability of the cells, they had a morphology like that of fibrocytes [ 22 ].…”

“…The presence of type II collagen (Col2A1) is related to the presence of healthy tissue, which, when hypertrophied after damage, begins to be replaced by type X collagen. On the other hand, metalloproteinase 13 (MMP- 13) is the most widely used marker of inflammation in the evaluation of cartilage, as it is the main protease in articular cartilage degradation [ 21 , 22 ].…”

“…Additionally, the hypertrophic condition also was observed OA condition as shown in Figure 5 . During progression of OA, chondrocytes convert to hypertrophic condition and disrupted arrangement of the chondrocytes [ 52 ]. This condition also elevates inflammation progression [ 53 ].…”
