A miniature integrated device for automated multistep genetic assays

Abstract: A highly integrated monolithic device was developed that automatically carries out a complex series of molecular processes on multiple samples. The device is capable of extracting and concentrating nucleic acids from milliliter aqueous samples and performing microliter chemical amplification, serial enzymatic reactions, metering, mixing and nucleic acid hybridization. The device, which is smaller than a credit card, can manipulate over 10 reagents in more than 60 sequential operations and was tested for the de… Show more

“…Multi-functional chips for sample pre-concentration, micro PCR amplification, and microchip electrophoretic separation have been demonstrated in several laboratories. [1][2][3] Currently, integration of sample processing steps into one system, to provide a complete sample-to-answer assay on the chip, is becoming the focus of development work.…”

“…Only a few prototype integrated DNA analysis devices that perform sample preparation, the analytical reaction, and the detection/ quantitation steps have been described (Table 1) (Woolley et al 1996;Burns et al 1998;Waters et al 1998;Brown 1999;Anderson et al 2000), and most do not integrate the sample preparation step of the analysis (Woolley et al 1996;Burns et al 1998;Waters et al 1998). This is because the sample preparation step is often the most difficult step in an assay, particularly for complex mixtures such as biological fluids.…”

“…Több sikeres DNS-hibridizációt is demonstráltak a chipen. Polikarbonát alapú chipeket is használnak a HIV-genom feltérképezéséhez [24], valamint üvegalapú eszközökben is ki lehet alakítani a DNShibridizációs vizsgálatokat [25].…”
