A minimally invasive, low-stress method for serial blood collection in aging mice

Abstract: Hematologic analysis is an efficient and valuable tool for real-time health monitoring and immune analysis in mouse aging studies. However, many frequently used blood sampling techniques in mice are incompatible with continuous monitoring with increasing age, as they may involve anesthesia, cause severe stress, or require a high volume of blood. This technical report describes a convenient relatively noninvasive procedure for counting white blood cells in C57Bl/6 mice by an optimized tail blood collection meth… Show more

“…The blood samples collected in EDTA were used to perform a manual count of white blood cells using Turk’s solution ( Bjorner and Zhu, 2019 ). A 100-cell differential leucocyte counts (DWBC) was performed manually using a blood smear stained with Romanowsky solutions ( Blumenreich, 1990 ).…”

“…Türk's solution consists of acetic acid, which is responsible for the hemolysis, and crystal violet to stain the remaining leukocytes, making them easier to see and count (16,33,34). Therefore, Türk's solution concentration test appeared to be an appropriate substitute to modify Knott's technique.…”

“…Specifically, blood was collected after a minimal puncture at the tail vein with a needle. 35 Individual mouse urine was aspirated and feces were collected after a gentle stimulus excretion and defecation on a clean disposable plastic container. 36 After the behavioral experiment, mice were fasted for 24 hours and weighed, and then sacrificed by cervical dislocation after femoral artery blood collection.…”
