Metal-based drugs have been developed in order to minimize side effects and/or to overcome intrinsic or acquired resistance to antitumor drugs available in medical clinics. In this sense, we are also looking for new pharmacological agents capable of curing or at least prolonging the survival of patients with tumors classified as incurable until then. Considering the need for rational production of new antitumor agents, this work describes the synthesis and structural characterization of new complexes of Pt (II) and Pd (II) of the type [M(L1)2(L2)2] (L1 = derived from 5-alkyl-1,3,4-oxadiazole-2-thione (5-alkyl-odztH) and L2 = triphenylphosphine) and of the type [M(L1)2L2] (L1 = derived from 5-alkyl-odztH or 5-amino-1,3,4-thiadiazole-2-thiol (5amino-tdztH) and L2 = 1,10-phenanthroline). The platinum and palladium complexes were characterized by elemental analysis, conductivity measurements, high-resolution electrospray ionization mass spectrometry (HRESIMS), infrared spectroscopy (FT-IR) and multinuclear NMR spectroscopy. Three complexes of the type [Pt(L1)2(PPh3)2] and one complex of the type [Pt(L1)2fen] had their crystalline structures determined by single-crystal X-ray diffraction. In all the complexes, the metal ion was coordinated with the two L1 ligands via a sulfur atom, in which two triphenylphosphine molecules or one phenanthroline molecule completes its coordination sphere. The molar conductivity values of 1.0 × 10 −3 mol L −1 solutions confirm that the complexes are non-electrolytes. The cytotoxic activity of the [Pt(heptyl-odzt)2fen] and [Pt(nonyl-odzt)2fen] complexes was evaluated in two tumor-cell lines (MCF-7 and MDA-MB-231), with the complex [Pt(5-nonil-odztH) 2 fen] being more active in both, since their IC50 values were lower. DNA interaction studies for both complexes reveled Kb values on the order of 10 4 M −1 , with no variation observed in positive or negative bands of circular dichroism (CD) spectra. In turn, in the fluorescence spectra of the Hoechst-DNA system, an observed reduction in fluorescence intensity indicated that these complexes interact with DNA through the minor groove.

Herein we report the synthesis and characterization of a new copper(I) complex with triphenylphosphine (PPh 3 ) and N-(2thiophenecarbonyl)-N'-(3-Cl, 4-F-phenyl)thiourea (HL), as ligands. The complex was characterized by vibrational (FTIR and FT-Raman) and multinuclear ( 1 H, 13 C { 1 H}, 31 P{ 1 H}) NMR spectroscopies. The crystalline structure of the complex was determined by single-crystal X-ray diffraction, confirming that a neutral binuclear compound, identified as [Cu(PPh 3 )(L-k 2 -N,μ-S)] 2 , was obtained. The anionic thiourea ligand coordinates to the metal through the sulfur and nitrogen atoms in an unusual bidentate k 2 -N,μ-S coordination mode. The complex is a dimer sited on a crystallographic center of symmetry. Each Cu 2 (μ-S) 2 cation bridges trough the sulfur atoms of two symmetry related thiourea moieties and is also coordinated to the nitrogen atom of the thiourea ligand and the phosphorus atom from PPh 3 coligand, forming a slightly distorted tetrahedral geometry. An intramolecular NÀ H⋅⋅⋅O=C hydrogen bond is observed in the anionic ligand, forming a six-membered ring that stabilizes the NÀ H thioamide group. Hirshfeld surface analysis shows that the molecules are connected by weak intermolecular contacts C⋅⋅⋅C, H⋅⋅⋅C and H⋅⋅⋅H which add to the stability of the crystalline packing. The in vitro cytotoxicity study of the complex indicates that it is more active against human lung carcinoma cells (A549), when compared to the free ligand.

The crystal structures of seven metal dihalide complexes with triphenylphosphine (PPh₃) and two with tribenzylphosphine (PBz₃) have been reanalyzed. The studied complexes include [FeCl₂(PPh₃)₂], [CoCl₂(PPh₃)₂], [CoBr₂(PPh₃)₂], [NiCl₂(PPh₃)₂], [NiBr₂(PPh₃)₂], [NiI₂(PPh₃)₂], [ZnCl₂(PPh₃)₂], [NiCl₂(PBz₃)₂], and [NiBr₂(PBz₃)₂]. A detailed geometric analysis revealed high isostructurality among several pairs. Ligand steric effects were assessed using cone angles and percentage surface coverage. Crystal packing is stabilized by weak C─H⋯π and C─H⋯X (X = Cl, Br, and I) interactions. Notably, [NiCl₂(PBz₃)₂] and [NiBr₂(PBz₃)₂] feature rare intramolecular C─H⋯Ni anagostic interactions. Reduced density gradient (RDG) and noncovalent interaction (NCI) plots confirmed weakly attractive C─H⋯π contacts in all complexes. Void‐volume analysis assessed the mechanical strength of the crystals, while pixel energy and energy frameworks clarified structural stability and dominant interactions. Density Functional Theory (DFT) calculations, along with QTAIM/NCIplot and NBO analyses, as well as electron density (ED) versus electrostatic potential (ESP) plots, were performed to better understand the C─H···Ni interactions and distinguish between agostic and anagostic interactions.
