A Modified Harlequin Stain For Cell Kinetics

Abstract: Lymphocytes cultured with 4 × 10‐4 m bromodeoxyuridine (BrdU) and stained by the harlequin procedure show wide variations in the colour and texture of their interphase nuclei. We demonstrate that we are able, by inspection, to identify four sets of nuclei: small dark nuclei (D) in cells that have not transformed in response to mitogenic stimulation; red nuclei (R) in transformed cells that have not synthesized DNA in culture; speckled nuclei (S)—red speckles on a bluish background—in cells that are synthesizin… Show more

“…Cell material and time are saved and sample fluctuations are reduced because aliquots of cell suspensions prepared for cytogenetic studies can be used for this test. A similar technique was proposed by Pincu et al (1986), but their method requires 120 mg ml 21 BrdU instead of the 6 mg ml 21 used in the present study. The validity of the method proposed by Pincu et al is limited because BrdU at concentrations §10 mg ml 21 is genotoxic in many cell systems (Morris 1991 and references therein) and exerts a strong inhibitory effect on cell proliferation (e.g.…”
