A modified histochemical technique to visualize acetylcholinesterase-containing axons.

Abstract: An improved histochemical method for light microscopic demonstration of acetylcholinesterase (AChE) has been developed. Axonal, dendritic, and perikaryal staining are well delineated, both in areas of low AChE content, such as cerebral cortex, and in areas of high AChE content, such as neostriatum. Axonal staining, including arborizations, stands out against a clear background devoid of diffuse reaction product.

“…Sections were then thaw mounted on gelatin-subbed glass microscope slides and stored at À801C until further processing. Sections were stained using an acetylcholinesterase staining protocol modified from Hedreen et al (1985). Briefly, sections were brought to room temperature and placed in a staining solution of 0.2 M Tris maleate pH 7.5, 0.1 M sodium citrate, 0.03 M cupric sulfate, 5 mM potassium ferricyanide, and 25 mg acetylthiocholine iodide for 75 min.…”

“…For AChE histochemistry, free-¯oating sections were post®xed by immersion in a 2.5% glutardialdehyde solution in 0.01 M PB saline (pH = 7.4) overnight at 4°C. Cholinergic ®bres were visualized by staining for AChE according to Hedreen et al (1985) using a silver nitrate intensi®cation procedure.…”
