A MODIFIED MINIMAL HEMOLYMPH-LIKE SOLUTION, HL3.1, FOR PHYSIOLOGICAL RECORDINGS AT THE NEUROMUSCULAR JUNCTIONS OF NORMAL AND MUTANTDROSOPHILALARVAE

Abstract: The hemolymph-like HL3 saline(Stewart et al., 1994)and standard saline(Jan & Jan, 1976)are two widely used bathing solutions for physiological recordings at the Drosophila larval neuromuscular junction. It has been established that longevity of larval preparations is better maintained in HL3 saline. However, HL3 can produce results that are inconsistent with previous findings in standard saline, particularly on temperature sensitivity and membrane excitability phenotypes. In wild-type larvae, the excitatory ju… Show more

“…Postfeeding third instar larvae crawling on the wall of food vials were chosen for recording. Dissections were performed in HL3.1 saline containing (in mM) 70 NaCl, 5 KCl, 4 MgCl 2 , 10 NaHCO 3 , 5 trehalose, 115 Sucrose, and 5 HEPES, at pH 7.2, with or without 1 mM CaCl 2 (Feng et al, 2004). [Ca 2ϩ ] o in recording saline varied in different experiments as specified.…”

“…Wandering third-instar larvae were dissected in HL3 (Stewart et al, 1994) for data collected at 0.4 mM, 0.6 mM, 1.0 mM and 1.5 mM [Ca 2+ ], or HL3.1 (Feng et al, 2004) for data collected at 0.1 mM [Ca 2+ ]. Recordings were performed as described previously (Howlett et al, 2008).…”

“…Tissues were then mounted and viewed on a Zeiss LSM 710 confocal microscope. For calcium imaging and fused atrial naturietic factor (ANF)-GFP imaging experiments, adult ring glands were dissected and placed in adult hemolymph-like (Feng et al 2004) solution containing 12 mM trehalose, which mimics replete levels (Broughton et al 2005). Following a 15-min incubation period, explanted ring glands were then viewed on a Zeiss LSM 710 confocal microscope.…”
