A Molecular Toolkit for Highly Insulating Tethered Bilayer Lipid Membranes on Various Substrates

Abstract: Tethered bilayer lipid membranes (tBLMs) are promising model architectures that mimic the structure and function of natural biomembranes. They provide a fluid, stable, and electrically sealing platform for the study of membrane related processes, specifically, the function of incorporated membrane proteins. This paper presents a generic approach toward the synthesis of functional tBLMs adapted for application to various surfaces. The central element of a tethered membrane consists of a lipid bilayer. Its proxi… Show more

“…Lipid monolayers were formed via self-assembly by immera͒ sion of an ultra flat gold substrate into a diluted solution of the anchorlipid DPhyTL ͑0.2 mg/mL͒, which was synthesized as described previously. 21,27,28 DPhyTL consists of two phytanyl chains that are coupled via a glycerol linker to a tetraethylene oxide spacer. The molecules are grafted to the gold surface by sulfur-gold bonds via a lipoic acid moiety.…”

“…C SC represents the space charge capacitance due to the spacer region combined with the capacitive effects of the electrochemical double layer at the gold interface. 21,24 All electrochemical experiments were performed in 0.1 M KCl unless otherwise noted. For the long time measurements, the solution has been replaced before each measurement, ensuring that the membrane did not dry out.…”

“…C SC represents the space charge capacitance due to the spacer region combined with the capacitive effects of the electrochemical double layer at the gold interface. 28,35 In the case of low monolayer resistances, an additional resistance attributed to the spacer region can be seen; therefore, an R͑RC͒͑RC͒ circuit has been used. 36 EIS data can be visualized in Bode plots ͑see Fig.…”
