A Moloney Murine Leukemia Virus-Based Retrovirus with 4070A Long Terminal Repeat Sequences Induces a High Incidence of Myeloid as Well as Lymphoid Neoplasms

Abstract: Retroviruses can be used to accelerate hematopoietic cancers predisposed to neoplastic disease by prior genetic manipulations such as in transgenic or knockout mice. The virus imparts a second neoplastic "hit," providing evidence that the initial hit is transforming. In the present study, a unique retrovirus was developed that can induce a high incidence of myeloid disease and has a broad host range. This agent is a Moloney murine leukemia virus (Mo-MuLV)-based virus that has most of the U3 region of the long … Show more

“…20 The injection of MOL4070LTR profoundly accelerated the onset of the disease in the Fbxl10 Tg group (median survival 5 274 days, P , .0001) ( Figure 1D). We also applied a chemically (N-ethyl-N-nitrosourea [ENU]) induced leukemia model to the same cohort, which enhanced leukemogenesis in Tg mice (median survival 5 210 days, P , .0001) ( Figure 1E).…”

“…19 Briefly, pups received a single intraperitoneal injection containing approximately 2 ϫ 10 6 viral particles between 3 and 5 days of age. Genomic DNA was extracted from the bone marrow and/or spleen of mice that developed AML, and the host-viral junction fragments were cloned and mapped as described.…”

“…We used the MOL4070LTR retrovirus (33) to perform RIM in Mx1-Cre, Kras G12D mice. Here we show that aberrant Ikaros expression due to viral integrations is a frequent early event and that somatic Notch1 mutations arise later and cooperate with oncogenic Kras in leukemogenesis.…”
