Objective: The main objective of the study was to investigate the effect of ethanol seed extracts of Nelumbo nucifera Gaertn (Nymphaeaceae) as antihyperglycemic, anti-hyperlipidemic and antioxidant activity in streptozotocin-induced diabetic rats.Methods: Diabetes was induced in wistar albino rats by administration of streptozotocin (single intraperitoneal dose of 60 mg/kg B. W). The ethanol extract of N. nucifera seed at a dose of 200 and 400 mg/kg body weight was administrated at a single dose per day to STZ induced diabetic rats for a period of 42 d. The different pharmacological parameters were evaluated. The effect of ethanol seed extract of N. nucifera on insulin, blood glucose, urea, creatinine, HbA1C, serum protein, albumin, globulin, serum enzymes, serum lipid profiles, lipid peroxides (LPO) and other antioxidant enzymes like catalase (CAT), glutathione peroxidase (GPx), superoxide dismutase (SOD) and reduced glutathione (GSH) were measured in the diabetic rats. Results:In the acute toxicity study ethanol seed extract of Nelumbo nucifera were non-toxic at 2000 mg/kg in rats. The ethanol extract of Nelumbo nucifera seed showed significant reduction in blood glucose (p<0.05), serum enzymes (SGPT, SGOT, ALP) (p<0.05), lipid parameters (TC, TG, VLDL, LDL) (p<0.05) except HDL and significantly increased insulin (p<0.05), HDL (p<0.05), GPx, GSH, SOD and CAT (p<0.05) at the dose of 400 mg/kg when compared with the diabetic-induced control. Conclusion:The present study suggested that the NNSE has significant (p<0.05) antihyperglycemic, antihyperlipidemic and antioxidant activity in STZ induced diabetic rats. These results clearly indicate that Nelumbo nucifera is effective against free radical-mediated diseases, thus replacing the synthetic ones.

Abstract:The present work was conducted to evaluate the effect of fortified pan bread with 5%, 10%, and 15% date pits powder on the chemical constituents, the nutritional value, and rheological characteristics of different dough samples using farinograph and on its sensory properties. The rheological properties of dough referred that there was an increase in water absorption in all blends compared to the control sample, and there was an increase in dough development, dough stability and degree of softening at 15% compared with 5% or 10% only. Pan bread fortified with 15% date pits showed the highest score in overall acceptability when compared to control pan bread and other various concentrations of pits. Twenty four adult male albino rats were divided into four groups. The first group (6 rats) fed on basal diet as a (negative control group). The second main group (6 rats) was injected with alloxan to induce hyperglycemia (positive control group) and then the rats fed on the same basal diet. The other 12 rats after being injected with alloxan were divided into 2 groups (3 & 4) and received basal diets containing fortified bread with 10% and 15% date pits powder for 45 days respectively. Biological evaluation was carried out by determination of body weight gain and food intake. At the end of the experimental period rats were sacrificed, blood samples were collected from the aorta then separate serum to determine glucose, insulin, HbA1c, total cholesterol and other lipid , and also to determine each of kidney and liver functions of experimental rats. Liver and Pancreas were removed to demonstrate histopathological observation. Our results showed significant reduction in the glucose, HbA1c, TC, TG, LDL-C and VLDL-C, as well as reducing hazards on liver and kidney functions compared with positive control group. Histopathological observation proved that the last group diet (with 15% date pits) looked like the negative control group. So date pits powder fortified bread is recommended to gain nutritional and healthy benefit to decrease the risk of diabetic diseases.

Abstract. The aim of this study was to examine the combined effect of ursolic acid (UA) and rosiglitazone (RSG) on metabolic syndrome in C57BL/6J mice. Upon feeding high fat diet (HFD) C57BL/6J mice developed obesity, insulin resistance, dyslipidemia and hypertension. The male mice were randomly divided into six groups, and fed normal diet, normal diet plus UA and RSG, HFD alone, HFD plus UA, HFD plus RSG, and HFD plus UA and RSG, respectively. HFD fed mice showed increase in body weight, elevated plasma glucose and insulin. Activities of gluconeogenic enzymes such as glucose 6-phosphatase, fructose 1,6-bisphosphatase increased while the activity of glycolytic enzyme, glucokinase, decreased in the liver along with glycogen content. Total cholesterol, triglyceride, low-density lipoprotein cholesterol and very low-density lipoprotein cholesterol and free fatty acid levels significantly increased in the plasma, whereas high-density lipoprotein cholesterol significantly decreased in high fat diet fed mice. In addition, both systolic and diastolic blood pressure was increased significantly. Combined treatment with UA and RSG improved the above parameters towards normality and pronounced more responses than UA or RSG lone treatment. The inclusion of UA in treatment with RSG may reduce the body weight gain, one of adverse side effect associated with the RSG-therapy.
