A New Rat Model of Cisplatin-induced Neuropathic Pain

Abstract: BackgroundChemotherapy-induced peripheral neuropathy is a major side effect of anti-cancer drugs, and our knowledge of its mechanisms is lacking. Several models for chemotherapy-induced neuropathy have been introduced. However, the outcomes of these models differ significantly among laboratories. Our object was to create a model of chemotherapy-induced neuropathy in rats with cancer.MethodsFemale Sprague-Dawley rats were used. Mammary rat metastasis tumor (MRMT-1) cells were implanted subcutaneously in rats. C… Show more

“…Cisplatin-induced sciatic nerve damage in the rat model has been proven in many studies [34]. Cisplatin has a molecular affinity for the peripheral nerves as they lack the vascular and lymphatic barriers, which make them more susceptible to the precipitation of the cisplatin debris [25,41].…”

“…Reported literature on the effect of cisplatin on thermal sensitivity currently shows a discrepancy in murine models. Although one study saw no difference in thermal hyperalgesia on hot plate studies (46), others demonstrate enhanced sensitivity to noxious heat following cisplatin, more so than oxaliplatin, treatment (43,47,48). Likewise, mechanical hyperalgesia has been documented in mice as measured by decreased pressure tolerance on von Frey filament tests.…”

“…Thus, patients with breast cancer receiving this drug class often suffer from neuropathy caused by medication side effects during treatment. However, this side effect can affect the patient for many years and sometimes indefinitely [35][36][37][38][39]. In our reported patient, mental neuropathy cannot be attributed to chemotherapy drugs, because the neuropathy had developed recently, but chemotherapy had finished three years earlier, and the patient also had no signs of sensory impairment during treatment.…”
