A New Sesquiterpene Ester from Celastrus orbiculatus Reversing Multidrug Resistance in Cancer Cells

Abstract: In a search for revertants of multidrug-resistance in cancer cells, a novel (1) and two known (2,3) sesquiterpene esters were isolated from the root of Celastrus orbiculatus. The structure of 1 was elucidated as 1 beta,2 beta-diacetoxy-6 alpha,9 alpha-bis(benzoyloxy)dihydro-beta-agarofuran. Compounds 1-3 partially or completely reversed resistance to adriamycin, vinblastine, and paclitaxel of multidrug-resistant KB-V1 and MCF7/ADR cells.

“…Subfraction F [3][4] was further purified by preparative HPLC (YMC-pack ODS-AM, 250 × 20 mm i.d., 85% MeOH in H 2 O) to afford compound 1 (t R = 29 min, 14.7 mg). Fraction F 5 (eluted by petroleum ether/acetone, 100:10 and 100:15), using the above MPLC (67−87% MeOH in H 2 O), provided 12 subfractions F 5-1 −F [5][6][7][8][9][10][11][12] , and the following purification of F 5-1 by the Computational Methods. Details have been previously reported.…”

“…-Celastrus paniculatus (Celastraceae) is an evergreen shrub distributed throughout Hengchun peninsula of Taiwan, India, and Malaysia [1]. The family Celastraceae is well known for producing dihydroagarofuran derivatives and alkaloids [2], some of which exhibit insecticidal [3], antitumor [4] [5], anti-inflammatory [6], multidrug-resistance (MDR) reversing [7] [8], and immunosuppressive [9] activities. Moreover, seed oil of C. paniculatus has been reported to improve memory [10] and intestinal complaints [11] [12], and display antioxidant [13], and hypolipidemic [14] effects.…”

“…The new compound 1 was isolated as amorphous, optically active powder ([a] MS and 13 C-NMR analyses. UV Absorptions at 231, 274, and 281 nm were similar to those of orbiculin A [7], and suggested the presence of aromatic moieties. The presence of ester C¼O groups was indicated by IR bands at 1746 and 1718 cm Table 1).…”
