A new technique for monoconidial culture of the most aggressive isolate in a given population of Bipolaris sorokiniana, cause of foliar spot blotch in wheat and barley

Abstract: We developed a new technique for monoconidial culture of the most aggressive isolate in a given population of Bipolaris sorokiniana, to facilitate the evaluation of spot blotch resistance in wheat and barley. Blotched portions of infected barley leaves were placed on a glass slide in a moist chamber for production of conidia by associated fungal hyphae. Conidia were collected separately and grown on water agar discs. Individual water agar discs having conidium growth were inoculated on barley leaves. The conid… Show more

“…SD40 is one of the most virulent isolate predominantly found in United States Great Plains areas and routinely used to screen breeding materials (provided by Dr. Shaukat Ali, SDSU). The isolate was originally derived from a single spore and method of its isolation and cultivation is well described by Kumar et al (2007) . This fungus is easily identifiable based on the color morphology and number of nuclei in lab conditions ( Shoemaker, 1959 ).…”

It is one of the major concerns in South Asia including China, Nepal, Pakistan, Bangladesh and the humid north eastern regions of India (Kumar et al, 2007;Chand et al, 2008;Singh et al, 2009;Vaish et al, 2011;Prasad et al, 2013). In addition, SB is also considered as a serious threat to barley production in the upper Midwest of the United States and the prairie provinces of Canada (Clark, 1979;Ghazvini and Tekauz, 2007).…”

“…Total 10 samples were drawn from each genotype and 10 isolates of each population group were characterized further. Pure cultures were established by single spore isolation technique [16]. Cultures were maintained at 25°C by transfer on PDA and were revived at monthly interval.…”
