A new tool for cloning and gene expression in Streptococcus pneumoniae

Abstract: A new replicon suitable for cloning and gene expression was successfully introduced into Streptococcus pneumoniae. The non-integrative lactococcal vectors pIL253 (higher-copy) and pIL252 (lower-copy), which are based on the promiscuous theta-replicating plasmid pAMβ1, were established in pneumococcus. The stability and the small size of these plasmids, together with the presence of a helpful multi-cloning site make them a useful genetic tool for gene expression in this bacterium. The functionality of the syste… Show more

“…Interestingly, infection with the strain expressing RNase R in trans even diminishes the survival rate. This is indicative of a direct role of RNase R in the pneumococcus pathogenicity, as, in this strain, RNase R levels are higher than in the wild type [37].…”

“…A reversion to higher expression levels was observed in the complemented RNase R strain. However, in this strain, the NanA transcriptional levels were increased relative to the wild type amount, which might be due to the augmented level of plasmid-expressed RNase R [37]. Together, these results are indicative of a role for RNase R in the modulation of this virulence factor.…”

“…Gene expression mutants were constructed using the medium-copy-number expression vector pIL253 (31). Both operons targeted by genetic engineering are absent from the genome of L. rhamnosus ATCC 53103, which could therefore be employed as the heterologous expression host.…”

“…Transcriptomic analysis was performed to better understand how RNase R affects gene expression in S. pneumoniae clinical isolate TIGR4. We compared the transcriptome between Δ rnr mutant previously constructed [10, 12] and the WT by performing RNA-Seq of total RNA. We found 257 transcripts that had high expression values in either strain and more than 0.5 of log 2 FC between the two strains (Table S3 in Supplementary Material).…”
