A nitric acid plant tissue digest method for use with inductively coupled plasma spectrometry

Abstract: A wet digestion method for plant tissue analysis for use with Inductively Coupled Plasma -Atomic Emission Spectrometry has been developed. This procedure uses only nitric acid and eliminates the use of perchloric acid and subsequently eliminates the danger of HClO 4 . explosions, and the problem of KClO 4 precipitation. Analysis of National Bureau of Standards plant materials -orchard, tomato, and spinach leaves -digested with nitric-perchloric acid or nitric acid compared very well with the NBS certified valu… Show more

“…Each yeast culture was inoculated into 300 ml of YPD medium and grown until an A 600 of 1.0 was reached. Yeast cells were harvested and washed and dried at 70°C and subjected to inductively coupled plasma spectrometry analysis (44). To determine the soluble iron concentration, cells were sonicated and broken with a Sonic Dismembrator (Fisher), and the intracellular iron content was examined with a QuantiChron TM iron assay kit (BioAssay Systems, Hayward, CA).…”

“…Total N was determined by the Kjeldahl digestion method [15]. Available soil P was determined according to Bray and Kurtz [16], while plant tissue P was analyzed by the wet digestion method [17]. Exchangeable Ca, Mg, and K were extracted using 1 M NH 4 OAc [18] and the cations in the extracts were determined by Atomic Absorption Spectroscopy (AAS).…”
