A novel and sensitive method for determination of amisulpride in human plasma by two‐dimensional liquid chromatography

Abstract: A novel and sensitive heart-cutting two-dimensional liquid chromatography with ultraviolet detection method (2D-LC-UV) was developed and validated for determination of amisulpride in human plasma. The 2D-LC system consists of a first dimensional ( 1 D) LC column and a middle transfer column as well as a second-dimensional ( 2 D) LC column. After simple protein precipitation, the sample was directly injected into the introduction valve of the 2D-LC system. The 1 D column, playing a role of primary separation an… Show more

“…After centrifuging at a high speed of 20000 rpm for 10 minutes, 1000 µL of supernatant was transferred into a 2 mL sampling bottle, 500 µL of which would be used for analysis. The 500 µL supernatant was analyzed using two-dimensional high-performance liquid chromatography with ultraviolet detection (2D-LC-UV) [13].…”

“…After centrifugation at 20,000 rpm for 8 min, 1000 μL of supernatant was transferred into a 2 mL sampling bottle, 500 μL of which was ready for analysis. The 500 μL supernatant was verified using two-dimensional high-performance liquid chromatography with ultraviolet detection (2D-LC-UV) [1]. The method Z score was satisfactory (0.04); the method was certified by the Laboratory of the Government Chemist in the UK.…”

“…The amisulpride steady-state plasma concentration of the patients was determined by two-dimensional liquid chromatography. The method was developed and validated in our lab (Qu et al , 2021). Amisulpride in 400 μl plasma was extracted by the addition of acetonitrile.…”
