A novel chloroplast localized Rab GTPase protein CPRabA5e is involved in stress, development, thylakoid biogenesis and vesicle transport in Arabidopsis

Abstract: A novel Rab GTPase protein in Arabidopsis thaliana, CPRabA5e (CP = chloroplast localized) is located in chloroplasts and has a role in transport. Transient expression of CPRabA5e:EGFP fusion protein in tobacco (Nicotiana tabacum) leaves, and immunoblotting using Arabidopsis showed localization of CPRabA5e in chloroplasts (stroma and thylakoids). Ypt31/32 in the yeast Saccharomyces cerevisiae are involved in regulating vesicle transport, and CPRabA5e a close homolog of Ypt31/32, restores the growth of the ypt31… Show more

“…Arabidopsis CPRabA5e (where CP represents chloroplast) is a chloroplast-localized protein found to regulate oxidative stress signaling. The role of CPRabA5e was further validated by Y2H screen where 13 proteins previously predicted to be involved in stress and localized in thylakoids and plastoglobules were identifi ed (Karim et al 2014 ). However, the exact mechanism of CPRab5e activity is not known yet.…”

“…However, in the case of most components of the putative chloroplast vesicle transport system, unequivocal experimental validation of their localization and function is still missing. So far, only two, the small plastid GTPases CPSAR1 and CPRabA5e, have been investigated in molecular detail [47][48][49].…”

“…The Rab GTPase CPRabA5e is a close homolog of the vesicle transport protein Ypt31/32 from yeast and has recently been shown to complement the ypt31/32 mutant [49]. CPRabA5e localizes to the chloroplast stroma and thylakoids, and an A. thaliana knock-out mutant displayed enhanced chloroplast vesicle formation at low temperatures and under oxidative stress [49].…”

“…16 Therefore spatial segregation could be an indication of false positive results, or is reflecting the possibility that similar proteins (like the identified ones) are present within the given organelle. Regardless, it is interesting to note that several of the proteins identified, using yeast two-hybrid with CPRabA5e as bait 2 or Interactome 3D, have relations to vesicles, thylakoid formation and/or GTP cycling.…”
