“…In contrast to other radical scavenging assays, a DPPH radical is stable and Belgium) containing 10% FBS, HEPES Buffer (10 mM), L-glutamine (2 mM), gentamicin (50 µg/mL), penicillin (100 U/mL), and streptomycin sulfate (100 mg/mL). As a robust and classical antineoplastic apoptogenic reference agent (El-Hamoly et al, 2017), cisplatin (1-200 μM) was recruited for comparison purposes (Sweidan et al, 2017 ;Alabsi et al, 2018;Mamdooh et al, 2019;AlKhalil et al, 2020;Hamdan et al, 2020 ;Kasabri et al, 2020;Al-Nuaimi et al, 2021 ;Shakoor et al, 2021;Haj Hussein et al, 2022;Hallaq et al, 2022;Khaleel et al, 2022;Qashou et al, 2022;Salih et al, 2022). The mechanism of reduction of cell viability was adopted so that Dose-response curves were plotted and values were expressed as percentage of control optical density and IC 50 values 50% inhibitory concentration were estimated by regression analysis (Piazzini et al, 2019).…”

“…Selectivity index (SI) is the term that describes the safety of tested drugs. It is calculated by dividing IC 50 value of tested compound on fibroblasts by the least IC 50 value of the same compound on any specific pathological cell line (Hoffmann, et al, 2011;Sweidan et al, 2017 ;Mamdooh et al, 2019;AlKhalil et al, 2020;Hamdan et al, 2020 ;Kasabri et al, 2020;Al-Nuaimi et al, 2021 ;Shakoor et al, 2021;Haj Hussein et al, 2022;Hallaq et al, 2022;Khaleel et al, 2022;Qashou et al, 2022;Salih et al, 2022).…”

“…MTT is colorimetric assay that is based on the reduction of a yellow tetrazolium salt (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide or MTT) to purple formazan crystals by the mitochondrial dehydrogenases of the metabolically active cells. [ 34 35 36 37 38 39 40 ] Viable cells namely MCF7 (5000 cells/well), PC3 (6000 cells/well), A375 (5000 cells/well), PANC1 (5000 cells/well), A549 (7000 cells/well) and CACO2 (11000 cells/well) were seeded in 96-well tissue culture plates for each cell line in high glucose DMEM (Bio Whittaker, Verviers, Belgium) medium supplemented as aforementioned cells were kept in a humidified 5% CO2 incubator (EuroClone, Italy) at 37°C for 24 h. Media was replaced subsequently in the 96 well plates with 100 μL fresh media and 100 μL of the different treatments to be incubated for 72 h in 5% CO2 incubator (EuroClon, Italy) at 37°C. Freshly, prepared MTT salt (3-(4,5-dimethylthiazol-2yl)-2,5- diphenyltetrazolium bromide) (5 mg/mL) was then added to each well to give a final concentration of 0.5 μg/mL.…”

“…Dose-response curves were plotted values expressed as percentage of control optical density and IC 50 values of 50% inhibitory concentration were estimated by regression analysis. [ 34 35 36 37 38 39 40 ] MTT bioassay was utilized also for growth inhibition by capsaicin, nano-form capsaicin and the most promising NA compounds (6-Hexyloxy NA, 6-Octyloxy NA, 5-Methyl NA, 6-Thien-2yl- NA, 5,6-Dichloro NA and 6-Amino NA) with different concentrations (6.25-400 mg/mL) on MCF7, PC3, CACO2, A375, PANC1, and A549 cell lines and Isonicotinic Acid (IsoNA) on PC3 cell line. For the viability reduction by combinations of NA compounds with capsaicin or nano-form capsaicin on MCF7, PC3, CACO2, A375, PANC1 and A549 cell lines was assessed with different concentrations.…”

“…Lipophilicity was assumed to ease and facilitate crossing of lipophilic cell membranes of cancer cell lines, unlike the hydrophilic drug vosaroxin. The idea of this work was based on previous work by our group involving new lipophilic FQ derivatives with noticeably promising antiproliferative activity against human colon and breast cancer cell lines (Arabiyat, et al 2016 a,b;Al-Nuaimi, et al, 2021). It was revealed that the FQ scaffold (6-fluoro-8-nitro-4-oxo-1, 4-dihydroquinoline-3-carboxylic acid) exhibited reasonable antiproliferative activity.…”
